Objective:To discussion the effect of BMSCs and chondrocytes are cultured on the basis of proportion of 3:1 as seed cells, composite modified fibrin gel scaffold, to induce the formation ofcartilage in adult rabbits, the possibility of repair of cartilage damage.Methods:Were killed by overdose anesthesia of 1 week old rabbits, separation and cultivation of BMSCs by whole bone marrow adherent culture method; separation of rabbit knee and elbow joint cartilage cells at the end of type 2 collagen enzyme digestion. Take the third generation and the first generation BMSCs articular cartilage cells, cell density and mixed by 3:1, then mixed with the modified fibrin gel scaffold composite.48 adult Japanese big ear rabbit full-thickness cartilage defect model with an electric drill in the bilateral knee femoral trochlea of intermediate in the manufacture of diameter 5mm,4mm deep,48 Japanese rabbits were randomly divided into three groups:the experimental group, the defect of injection improved fibrin gel and seed cell compound; control group. Defect injection improved fibrin gel; the blank group, the defect of exclusion. Specimens for 12 weeks in 6 weeks after surgery, after the repair from the gross observation, histological grading, Wakitani HE staining, immunohistochemical staining and GAG content detection, evaluation of rabbit articular cartilage defects.Results:The observation group:gross observation and A group (experimental group) after 12 weeks of defect is new repaire tissue coverage, repair tissue similar to articular cartilage and cartilage tissue damage around, blurred boundary; group B (control group) in the mend of tissue to mend cartilage damage can,t be whole transparent, less the content and the defect of cartilage, cartilage tissue around the boundary is clear; C group (blank group) repair effect is poor, the scar tissue repair.Conclusion:1,The modified fibrin gel scaffold has good biocompatibility, degradation rate is appropriate, do not induce inflammatory reaction, in which the normal chondrocytes proliferation and secretion of matrix, as injectable cell scaffold is an ideal choice.2, the cell scaffold composite cartilage cells and the third generation of BMSCs, the construction of first improved fibrin glue generation by way of injection of perfect fill cartilage defect,12 weeks after the operation, can better repair of articular cartilage defects.
