Circadian rhythm is part of chronomedicne research.Circadian rhythm is also an important part of heaven corresponding research.From the perspectiv e of Chinese and Western medicine, the research of circadian rhythms is ne cessary.Melatonin and 5-hydroxytryptamine are important indicators in circadian r hythm.In the circadian rhythm, serotonin and melatonin match the function. Melatonin and 5-hydroxytryptamine are the main hormone of pineal gland r egulating the outer periphery physiological activity of circadian rhythm. Mel atonin and 5-hydroxytryptamine is an important indicator of circadian rhyth m research.Time series analysis method is suitable for interdependence between befor e and after data.Time series model can be quickly extracted from the overal 1 characteristics of time series data.Heaven corresponding research have the analysis of many rhythm indicators.Time series analysis methods have been applied to try the analysis of melatonin and 5-hydroxytryptamine.Hope to co ntribute the analysis and research of heaven corresponding index.Combining the characteristics of melatonin and 5-hydroxytryptamine.time s eries analysis method is suitable for analysis of melatonin and 5-hydroxytry ptamine secretion.This paper analyzes the circadian secretion of melatonin a nd 5-hydroxytryptamine by time series analysis to study the effect of interv ention on circadian rhythm in mice.1 ObjectThis paper attempts to analyze 5-hydroxytryptamine and melatonin circadian by time series analysis method to explore circadian rhythm research.Application of time series analysis method provide a reference for the analysis of rhythm indicators in heaven corresponding research.2 Method2.1 First experiment Effect of light on time series of 5-hydroxytryptamine and melatonin in mice2.1.1 Design and grouping of experimentExperiental animals are Kunming (KM) female mice,supplied by Beijing Vital River company.Kunming (KM) mouse number is 288, divided into two groups. Each group number is 144.(1) Control group:Mice were fed with normal diet under natural light condition.(2) Light group:Mice were fed with normal diet and use electronic timers to illuminate 18:00 to 24:00 daily.Each group randomly selected 6 to blood at 0 o’clock.Each group randomly selected 6 to blood at 2 o’clock.Continue to operate every 2 hours until each finished.The blood samples were taken by 3500rpm/15min-20min centrifuge to separate the serum.Serum into tubes and stored at low temperature (-70 ℃).Serum melatonin and 5-hydroxytryptamine levels measured by enzyme-linked immunosorbent assay.2.1.2 statistical analysis methodThe average of melatonin and 5-hydroxytryptamine as time series is analy zed. According to the time sequence diagram showing the characteristics of the timeseries, fitting model to establish melatonin and 5-hydroxytryptamine model.The model effect of fitting is analyzed.2.2 Second experiment The effect of light and drug on 5-hydroxytrypta mine and melatonin secretion2.2.1 Design and grouping of experimentStudy the secretion change from 0 to 6 outside illumination time.The experimental design was two-level two-factor repeated measurements.Two factors are light and drug.Two levels of illumination are light and non-light.Two levels of drug is drug and drug-free.Experiental animals are Kunming (KM) mice,supplied by Beijing Vital River company.Kunming (KM) mouse number is 24, divided into four groups..Each group number is 6.(1) Control group:Mice were fed with normal diet under natural light condition.(2) Drug group:Mice were fed with containing 0.25% of kidney herbs (Qi Rong Yi Jing particles) under natural light condition.(3) Light group:Mice were fed with normal diet and use electronic timers to illuminate 18:00 to 0:00 daily.(4) Light drug group:Mice were fed with containing 0.25% of kidney herbs (Qi Rong Yi Jing particles) and use electronic timers to illuminate 18:00 to 24:00 daily.Continuous blood every 2 hours after molding.The blood samples were taken by 350()rpm/15min-20min centrifuge to separate the serum.Serum into tubes and stored at low temperature.Serum melatonin and 5-hydroxytryptaminc levels measured by enzyme-linked immunosorbent assay.2.2.2 statistical analysis methodMelatonin and 5-hydroxytryptamine of 0 to 6 o’clock conduct repeated measurements analysis of variance to analyze the role of additional light, light and drug interaction.Each specific time point is analyzed too.2.3 Third experiment Time series analysis of 5-hydroxytryptamine and melatonin in different sex mice2.3.1 Design and grouping of experimentExperiental animals are Kunming (KM) mice,supplied by Beijing Vital River company.Kunming (KM) mouse number is 288.divided into two groups by sex. Drug has significant impact on melatonin secretion (P<0. 05) Light has significant impact on 5-hydroxytryptamine secretion (P<0.05).11 luminate time is 18:00 to 0:00, light has significant impact on melatonin se cretionat 2 o’clock and 4 o’clock (P<0.05).Drug,light and drug interact ion have significant impact on 5-hydroxytryptamine secretion at 0 o’clock (P<0.05)3.3 The different gender modeling effect of melatonin and 5-hydroxytryp tamineTime series model of melatonin and 5-hydroxytryptamine is simple season al type model. Melatonin and 5-hydroxytryptamine model fitting effect of fe male is good, smooth R are all above 80%. The model is good. The smoot h R of 5-hydroxytryptamine is 67.9%.The model fitting need to continue e xploration.3.4 Time series models integration still need to explore.The smooth R of model integration are all below 80% by direct modeling. Time series models integration still need to continue to explore model fitti ng.4 Conclusion4.1 Light influence secretion rhythm and its continuous functionCircadian rhythm of the control group, the light group are still.The light group has little rhythm of 12 hours.The illuminate time is 18:00 to 0:00. Li ght has still have significant impact on melatonin secretion at 2 o’clock an d 4 o’clock. Light has a continuing effect on melatonin secretion.4.2 Drug influence exist difference at time pointsDrug influence has significant impact on melatonin of groups. Drug still has significant impact on melatonin secretion at 0 o’clock. Drug has no si gnificant impact on other points.4.3 The advantage of time series analysis applied in the circadian rhyth m researchThe type of time series model helps to analyze the overall production ch aracteristics. From the perspective of overall, groups are contrasted under di fferent experimental conditions. Provide the basis for positioning circadian r hythm study time.Provide the reference for rhythmic indicators analyses and studies in heaven corresponding research.
