Objective:To analysis changes of glucagon (GC), free fatty acid (FFA) about different stages of patients with type2diabetes in fasting, OGTT, L-Arg experiments. To further discusses the relationship between GC, FFA and blood glucose (BG), ulin0, ulin homeostasis model (HOMA-β), glucose metabolism, and hormone secretion curve of indicators also.Methods:Four studying groups, including T2DM group of45cases, IGT group of28cases, IFG Group22cases, NGT group30cases, to carry out OGTT respectively, and in the second day to carry out L-Arg test. Under fasting state, to detect blood glucose(FBG), ulin (F), glucagon (FGC), FFA, to calculate HOMA-β, ulin sensitivity index (ISI), HOMA-IR in the different group respectively. In OGTT, the30min,60min,90min and120min’s BG, INS, GC, was detected and to calculate the area under the curve of BG, INS, GC(AUCBG、AUCINS、AUCGC). In L-Arg experiments, the2min,4min and6min’s BG, INS, GC, was detected and to calculate the area under the curve of BG, INS, GC(AUCBG、AUCINS、AUCGC). Detecting BG, FFA’s level was in the RA-1000automatic biochemical analyzer, detecting, GC’s level by radioimmunoassay determination, the kit purchased from the Beijing titute of Atomic Energy.Results:There are obvious overall difference of FFA and FGC in3groups, circumstances of FGC is T2DM>IGR>NGT, circumstances of FGC is T2DM>IGR>NGT. FGC are negatively correlated with HOMA-β and ISI, and positively correlated with HOMA-IR in each of3groups. In the all subjects of103cases, FGC are positively correlated with FBG, HOMA-IR, no correlation with F and negatively correlated with HOMA-β, ISI. In the all subjects of103cases, FFA are positively correlated with FBG, HOMA-IR, and negatively correlated with F, HOMA-β, ISI. And it is positive correlation in T2DM group and IGR group, but it is no correlation in NGT group. There are obvious overall difference of GC’s level in3groups at the OGTT and L-Arg experiments. Circumstances of area under the curve of GC performances is T2DM>IGR>NGT, and circumstances of area under the curve of FFA performances is T2DM>IGR>NGT, AUCGC are significantly negatively correlated with with AUCBG, and significantly negatively correlated with AUC. Conclusion:In T2DM and IGR patients, glucagon and free fatty acid’s level is higher obviously than normal. Their level is positively correlated with blood glucose, HOMA-IR, and negatively correlated with, HOMA-β,ISI. In T2DM and IGR patients, there are positive correlation between glucagon and free fatty acids, and this links may exist independently.
