Effect Of Taohong Siwu Decoction On Notch1 Content In Rabbit Skin Transplantation Model

Objective:This study established Rabbit graft model.By Taohongsiwu decoction,to observe the general situation of anastomosis area,vascular density and the variation of Notch1. Evaluating the effection of different does Taohongsiwu decoction on the skin grafts.Methods:75rabbits, adaptive feeding for1weeks after weighing, then according to the weight from light to heavy sequence number, and finally apply the randomly number table method to divide them into5groups, named Taohongsiwu decoction low-dose group,middle-dose group,high-dose group, model group and sham operation group.There were15rabbits in each group. After modeling were administered, and in postoperative2d,5d,8d three time, randomly select five rabbits of each group to be sacrificed and cut the graft skin.Then detect the serum protein and mRNA content of Notch1by ELISA and RT-PCR assay.Results:(1)The microvessel density of Taohongsiwu decoction group is significantly better than the model group (P<0.05), the difference is statistically significant. The serum protein and mRNA content of Notchl in Taohongsiwu decoction group is higher than the model group (P<0.05), the difference is statistically significant.(2) The vascular density and Notchl content of Taohongsiwu decoction middle-does and high-dose group is significantly higher than low-dose group (P<0.05), the difference is statistically significant.(3) Compared the vascular density and Notchl content of Taohongsiwu decoction high-dose group with the middle-dose group, the difference is not statistically significant.Conclusion:After Rabbit skin graft,Taohongsiwu decoction can significantly improve and heighten histopathological grading, increase the density of blood vessels, improve Notchl levels, promote the survival of the grafted skin and angiogenesis. Compared with Taohongsiwu decoction low-dose group,the effect of high-dose group and middle-dose group are better.
