Purpose:Observation of the kidney of diabetic nephropathy rats aboutexpression of nephrin, VEGF, study on Chinese medicine YiTangKang about effectof nephrin mRNA gene and protein, VEGF mRNA gene and protein, to investigatethe mechanism of yitangkang about reduced urine protein, kidney protection。Material and method:60clean grade male SD rats, weight220±20g,12weeksof age, a week’s adaptive feeding, randomly selected10as normal controlgroup,another50intraperitoneal injection STZ to establish diabetes model.2weeks carrying DN model, according to the blood glucose values were randomlydivided into five groups,10rats in each group. Including:The model controlgroup, preventive group, low dose group of Chinese medicine, Chinese medicinehigh dose group, western medicine group. Each group according to thecorresponding drug and dose to administered. Lavage6weeks, detection of24hurine protein, serum creatinine, blood urea nitrogen, RT-PCR and Western blotto detect the expression of nephrin mRNA gene and protein, VEGF mRNA gene andprotein。Results:1.In the model groupwere,the serum24h urine protein, serum creatinine, bloodurea nitrogen are higher than that in normal control group, compared with themodel group, prevention group, low-dose group, high-dose group of ChineseMedicine The24h urine protein, serum creatinine, urea nitrogen levels decreasedsignificantly(p﹤0.01,p﹤0.05)。2. The model rats kidney tissue expression nephrin was lower than the controlgroup(p﹤0.01),compared with the model group, prevention group, low-dose group,high-dose nephrin in renal tissues of rats expression significantly increased (p﹤0.01),compared with the prevention group,low-dose group, high-dose nephrinin renal tissues of rats expression significantly decreased(p﹤0.01), comparedwith the low-dose group, prevention group and high-dose nephrin in renal tissuesof rats expression significantly increased(p﹤0.01)。3. Compared with normal control group was significantly increased VEGFexpression in renal tissue of rats in model group(p﹤0.01),compared with themodel group, prevention group, low-dose group, high-dose VEGF in renal tissuesof rats expression was decreased(p﹤0.01), compared with the prevention group,low-dose group, high-dose VEGF in renal tissues of rats expression significantlydecreased(p﹤0.01), compared with the low-dose group, prevention group andhigh-dose VEGF in renal tissues of rats expression significantly increased(p﹤0.01)。Conclusion:1. Traditional Chinese medicine of yitangkang reduce the24h urine proteinexcretion, reduce serum creatinine and urea nitrogen levels in rats。2. Traditional Chinese medicine of yitangkang promote rat kidney nephrin andnephrin mRNA expression,and may by maintaining enough cells and the integrityof the glomerular filtration barrier, to play the role of protecting kidney。3. Compared with the prevention group,low-dose group, high-dose nephrin in renaltissues of rats expression significantly decreased, prompting Yi Tang Kang earlyintervention may have better protective effect on renal podocyte DN modelrats.Nephrin expression in the kidney of the high dose Chinese traditionalmedicine group DN rats increased significantly, these results suggested thathigh concentration of yitangkang may be greater on renal podocyte protectionsignificance of DN model rats4. Traditional Chinese medicine of yitangkang inhibition of rat kidney VEGF,VEGF mRNA expression,it may is one of the target of prevention and treatmentof DN。5. Prevention of renal VEGF expression of group DN rats compared with traditionalChinese medicine low dose group, high dose group decreased significantly, PromptYi Tang Kang early intervention may have better protective effect on renalfunction of DN rats.VEGF expression in the kidney of the high dose Chinesetraditional medicine group DN rats increased significantly, these results suggested that high concentration of yitangkang may be greater protection tothe significance of kidney of DN rat model.
