"Eseye", a Tibetan medicine customarily used by folks in Ganzi, be derived from the dry leaves of Malus toringoides (Rehd.) Hughes, Malus transitoria (Batal.) Schneid.. Firstly recorded in Jing Jing Ben Cao material medica, it has been well used to treat indigestion and liver diseases. It has a long history of use in Ganzi, but lackded legal standards. Fristly, this article through correlation research between the chemical compositions and efficacy of, definited that the index compositions are active ingredients of this medicine, and studied the quality standard research.The main results were as follows:1. Correlation research between the chemical compositions and efficacyOn the basis of preliminary extraction and separation, phloretin, quercetin, phloridzin isolated from theleaves of Malus transitoria (Batal.) Schneid., are of over98%by HPLC.For the first time, through the study antioxidant activity of the leaves, fruits, flowers from original plants by DPPH, they all have certain antioxidant activity. Especially, the leaves and flower, their antioxidant activity were significantly higher than the fruit. Combining with the content of chemical composition and the antioxidant activity, contribution rates of phloretin and phloridzin in the antioxidant activity of medicinal materials are9.51%,2.88%, but quercetin0%. So, phloretin and phloridzin can be used as index compositions used in content determination, evaluation quality of Eseye.For the first time, it was studied the effect on protect liver experimental research of Eseye and its chemical composition. Different doses of Malus toringoides(Rehd.)Hughes, Malus transitoria (Batal.)Schneid., phloretin, phloridzin have protective effect on liver injury caused by CC14. Except the MDA level, the ALT, AST, SOD level, Eseye were better than Ganhuangcao, phloretin and phloridzin were better than bifendate. Eseye are with high dose (2g/kg) best, and phloretin in optimal dose (10mg/kg) best and phloridzin in low dose (5mg/kg) best. According to determine the best dose, the daily dosage of Eseye is3.3~13.2g, consistent with the compendium of basic, and phloretin, phloridzin are the active ingredients.2.Quality standard researchOn the basis of previous research, germplasm origin, morphological, microscopic, TLC, examination, extract, content determination of22batch Eseye were studied. TLC identification method in chloroform-methanol-formic acid (8:2:0.1) as developing agent, phloretin as reference substance, was established and has a good reproducibility. Content of flavonoids is determined by UV, phloridzin as reference substance, methanol as solvent extraction,. Content of phloretin is determined by HPLC method,40%ethanol as extraction solvent. Chromatographic condition was stable, with good reproducibility.Safety evaluation studY at the same time, Eseye, its LD50of is not less than15g/kg, belongs to the non-toxic substances.Through the above research, in terms of morphological, microscopic and content determination, leaf and leaf bud of Eseye are prescribed respectively. To develop a system, scientific Eseye quality standard, in order to ensure the safety and efficacy of Eseye the in Tibetan area, this provides scientific basis for effectively.
