Objective:The present study was to establish the non-infective fever rat model, to observe the content changes of cAMP (cAMP) and protein kinase A (PKA) in serum and hypothalamus tissues of fever rats and normal rats, to compare the relevant indexes changes of Qingresanyujiedu decoction and western medicine naproxen, to discuss the mechanism of treatment of cancerous fever with Qingresanyujiedu decoction. Methods:We select42SPF level SD rats with6weeks of age and randomly divided into4groups:①The saline control group (normal group, n=6);②The fever model group (model group, n=12);③The Qingresanyujiedu decoction group (treatment group, n=12);④The western medicine naproxen treatment group (control group, n=12); Model group, Chinese medicine group, control group rats subcutaneous injection of2,4-Dinitrophenol to induce endogenous fever model, the normal group not injection. Model group, Chinese medicine group, control group were intragastric administration after a successsful molding. Model group to saline model group, Chinese medicine group to Qingresanyujiedu decoction, the control to the naproxen sodium chloride solution. Modeling after2hours, we measure and record body temperature of each group rats. All groups were collect blood after putting rats to death. The hypothalamus tissues were removed, frozen, fixed in liquid nitrogen immediately and we were waiting for testing. According to the manual operation, the cAMP, PKA content in the serum and the hypothalamus tissues were detected respectively by using enzyme-linked immunosorbent method. Results:1. The rats body temperature was compared before and after building the model among each group. After injecting2,4-2nitro phenol2hours, Chinese traditional medicine group body temperature was (39.02±0.23)℃, the control group was (38.74±0.21)℃, model group was39.57±0.38)℃and blank group was (37.41±0.38)℃. By analysis of4groups variance, the temperature of Chinese traditional medicine group and control group is lower than model group (P<0.05), it has a statistically significance.2. The cAMP content was compared in serum and the hypothalamus tissues of rats after using drugs. The statistical analysis results of cAMP content between the three groups was P<0.05, it has statistically difference between the two groups, serum cAMP content of Chinese medicine group was obviously lower than the control group, the Chinese medicine group and the control group is lower than the model group. The statistical analysis results of hypothalamus cAMP content between the three groups was P<0.05. It has statistically difference between the two groups, the hypothalamus cAMP content of Chinese medicine group was obviously lower than the control group, the Chinese medicine group and the control group is lower than the model group. That Chinese medicine curative effect is superior to western medicine and traditional Chinese medicine and western medicine has curative effect.3. The PKA activity was compared in serum and the hypothalamus tissues of rats after using drugs. The statistical analysis results of PKA content between the three groups was P<0.05, Chinese medicine group and control group have statistically difference between with model group; and serum PKA content of control group was lower than Chinese medicine group, there was no statistical difference between the two groups. The statistical analysis results of hypothalamus PKA content between the three groups was P<0.05. Chinese medicine group and control group have statistically difference between with model group; and hypothalamus PKA content of control group was lower than Chinese medicine group, there was no statistical difference between the two groups. Traditional Chinese medicine and western medicine has curative effect. Conclusion:1. Qingresanyujiedu decoction can significantly reduce the body temperature of EP induced fever rats.2.The cAMP content and PKA activity in serum and hypothalamus tissues of EP induced fever rats were significantly increased than before and after modeling, the second signaling pathways (PKA system) play an important role in the process of fever.3.Qingresanyujiedu decoction can decrease cAMP content and inhibit PKA activity in serum and hypothalamus tissues of EP induced fever rats, it explains that one of the compound decoction effect mechanism is related with adjusting the second messenger pathways and decreasing the body’s cAMP content, PKA activity, the hypothalamus may be one of the targets.
