Effect Of Tongsheng Tongbi Paste On Expression Of MMP-1 And MMP-3 In Degenerated Articular Chondrocytes Of Rabbit Knee Osteoarthritis

Objective:To observe the effects of Dieda Tongbi emplastrum on MMP-1(matrix metalloproteinase-1) and MMP-3(matrix metalloproteinase-3) expression of the rabbit model of knee osteoarthritis,and explore its action mechanisms,thus provide the experimental support and theoretical for its further clinical application.Method:Randomly divide613-4months old,1.5-2.5kg healthy New Zealand purebred-rabbits were randomly divided into Blank group(A), Sham-operated Group(B),Modeling group(C),Dieda Tongbi emplastrum group(D),Gutong emplastrum group(E),12rabbits each group,and the other one is model rabbit of improved Hulth molding method.No treatment in Group A,but C,D and E were built by improved Hulth molding method, Group B were opened the joint cavities without any treatment and sutured directly.5weeks later,we got the models successfully,and then entered the stage of experimental treatment.No treatment in Group A and B,Group C were fixed with tape, Group D were applied with Dieda Tongbi emplastrum, Group E were applied with Gutong emplastrum.Tapes,Dieda Tongbi emplastrum,Gutong emplastrum were cropped to the size of2cm*4cm by calculation,which were fixed on the right knee one day at a time,removed them after8hours,applying for4weeks.5weeks later,observed the changes of rabbits behavior,changes under x-ray,changes of joint cartilage, MMP-1and MMP-3expression in joint cartilage.Results:1.All the experimental rabbit survived to the observing time without any allergy;2.The right knee joint radiography examination of Group D and Group E showed improvement after treatment compared with Group C.3.Observation of articular cartilage:â‘ Visual rating:Group D and E got lower score than Group C(P<0.05).â‘¡The electron microscopy of articular cartilage score:Group D and E scored lower than Group C(P<0.05). â‘¢. MMP-1and MMP-3test:MMP-1and MMP-3expression in Group C,D and E was significantly increased(P<0.01) compared with Group A,and their expression in Group D and E were reduced compared with Group C(P<0.01).Conclusions:1.Rabbit KOA models could successfully copied by Hulth modeling method;2.MMP-1and MMP-3expression in Synovial fluid of KOA models were higher than normal group, which proved that they were related with KOA;3.Dieda Tongbi emplastrum and Gutong emplastrum have therapeutic effect on KOA however not to cure KOA, which are capble of delaying the course of KOA,and has no statistical difference between them.
