Purpose:Discussion the inhibitory effect of BZYQD on human lung adenocarcinoma cell line A549 in nude mice and its mechanism, provide experimental evidence for clinical practice.Material and method:Inoculated Balb/c nude mice with A549 cells to establish a nude mice model. according to random number table, randomly divided 50 mice into control group, BZYQD low-dose group, BZYQD middle-dose group, BZYQD high-dose group, DDP group, counted from the first day of the start of the administration, coverage after 15 days of intervention, remove the solid tumors of nude mice and the weight them.hematoxylin-eosin (HE staining) was used to detect the morphological shape changes of mice’splenic white pulp by using stereology;immunohistochemistry SABC were used to detect the the expression of BZYQD on Caspase-3 protein of mice’spleen cells;one-step RT-PCR detect the expression of BZYQD on Caspase-3 mRNA of mice’spleen cells.Results:Tumor growth in nude mice in each experimental group, compared with t e control group,tumor weight decreased in BZYQD low-dose group,BZYQD medium d ose-group, BZYQD high-dose group and DDP group(p<0.05), the inhibition rate displa y BZYQD and DDP all have the effect of significantly inhibited tumor growth; comp ared with DDP group, tumor weight of BZYQD low-dose group,BZYQD medium-dose group and BZYQD high-dose group have no difference (p>0.05); tumor weight of B ZYQD low-dose group,BZYQD medium-dose group and BZYQD high-dose group hav e no difference (p>0.05)by pairwise comparisons; after hematoxylin-eosin staining, ster eological method to observe and calculations show that, compared with control group, BZYQD low, medium and high-dose groups,the relative area of white pulp of the sple en of nude mice was significantly increased (p<0.05),comparison between groups of B ZYQD low, medium and high-dose groups have no difference (p>0.05); immunohistoc in nude mice spleen of BZYQD low, medium and high-dose groups was significantly increased (p<0.05), comparison between groups of BZYQD low, medium and high-do se groups have no difference (p>0.05);one-step RT-PCR showed that,compared with th e control group, expression of Caspase-3 mRNA in nude mice spleen of BZYQD lo w, medium and high-dose groups was significantly increased (p<0.05),comparison bet ween groups of BZYQD low, medium and high-dose groups have no difference (p>0. 05).Conclusion:1. BZYQD has inhibitory effect on human lung adenocarcinoma cell line A549 in nude mice similar with the use of DDP,and this effect has non-dose-dependent;2. BZYQD can increase the relative area of white pulp of the spleen of nude mice, and this effect has non-dose-dependent;3. BZYQD can upregulate expression of Caspase-3 protein and mRNA in human lung adenocarcinoma cell line A549 in nude mice spleen,and this effect has non-dose-dependent;4.The inhibitory effect of BZYQD on human lung adenocarcinoma cell line A549 in nude mice, BZYQD may improve immune function by upregulate the expression of Caspase-3 protein and mRNA in nude mice spleen cells.
