Lepidium latifolium L., a perennial herbaceous plant, is a member of the genus Lepidium L. of the Brassicaceae family. As a rare and precious medicinal plant, It has traditionally been used as medical plant for thousands of years, which could be referred to Sheng Nong’s herbal classic. In this research, extracts with different polarity from Lepidium latifolium L. have been studied for the promotion of gastrointestinal pharmacodynamics. The aim of this research is to lay the theoretical basis for the decelopment of gastrointestinal pharmacodynamics medicine with chinese-owned intellectual property and for the better use of this medical plant.Ethanol, petroleum, ethyl acetate, n-butanol(with the concentration of 75%) were used as the solvent for the liquid-liquid extraction of the crude extract. Domperidone was taken as positive control medicine, meanwhile, atropine was acted as the inhibitor. Gastric emptying, intestinal propulsion, voltage amplitude of gastric myoelectrical activity had been tested in turn on mice and rabbits. The contractile tension of duodenum, the mid and distal small intestine, the contents of MTL in plasma, GAS in serum of had been measured on mice and rabbits as well. In addition, the experiment data were dealed with by SPSS17.0 to analyze and study the pharmacodynamic effect of extracts with different polarity from Lepidium latifolium L. The main achievements and conclusions are as follows.1 The extraction process parameters were optimized by orthogonal experiment on the basis of single factor tests. The optimum conditions for extraction are as follows: liquid-solid ratio 12:1,extracting temperature 80 ℃ and extracting time 1h.2 Compared with the blank control group, the ethyl acetate extract could obviously accelerate the gastric emptying and improve the gastric emptying of mice which resisted the inhibition of atropine(P<0.01). At the same time, the n-butanol extract could strengthen intestinal propulsion and improve intestinal propulsion of mice which resisted the inhibition of atropine significantly(P<0.01). It showed no significant difference compared with positive control group(P> 0.05).3 Ethyl acetate extract could rise the contents of MTL in plasma, GAS in serum of mice(P<0.01). It also could keep the level of MLT in plasma, GAS in serum of mice which resisted the inhibition of atropine simultaneously(P<0.01). There were no significant difference compared with positive control group(P> 0.05).4 Ethyl acetate fraction could enhance the voltage amplitude of gastric myoelectrical activity of rex rabbit. No significant difference has been detected in frequence and other control group.5.The fraction of ethyl acetate extraction could enhance the contractile tension of duodenum, the mid and distal small intestine of rex rabbit in vitro dramatically(P<0.05). No significant difference has been occurred in frequency and other control groups(P>0.05).The results of our studies indicated that the water extract from Lepidium latifolium L., n-butanol fraction and Ethyl acetate fraction all could have the function of promoting the motion of gastro-intestines. Ethyl acetate fraction showed the best bioactivity about gastrointestinal motility and it demonstrates that the biological constitutents of Lepidium latifolium L. exist in ethyl acetate extract mainly. Thus, the research on chemical constitutents and pharmacology of ethyl acetate extract in Lepidium latifolium L. is urgent and necessary in the future.
