Purpose:Osteoarthritis(abbreviated as OA),is one of clinical common joint disease. It is also called bone hyperplasia or degenerative joint disease in clinical. The main characteristic for the occurrence of chronic progressive articular cartilage degeneration and the subchondral bone sclerosis remodeling. OA,which ranks second only to the cardiovascular disease in adult disability disease,can affect quality of life in elders and cause economic loss,as one of the major diseases lead to long-term disability. As the world population aging,there is an upward trend in the incidence of the disease. Drynaria is is a genus of ferns in the family Drynaria fortune. The dry mature fruit of drynaria is the commonly used traditional Chinese medicine to strength kidney function since the ancient times. This medicine is recorded in Chinese pharmacopoeia as traditional Chinese medicine. Modern pharmacology study shows that effect of drynaria includes the expansion of the coronary arteries,antiBacterial,anti-aging,two-way adjustment on the smooth muscle,estrogen-like effects as well as anti-tumor and other pharmacological activity. Through experimental research,observation of the synovial tissue matrix metalloproteinases on osteoarthritis in rabbit model of-3(MMP-3),tumor necrosisfactor-α(TNF-α),interleukin-6(IL-6),superoxidedismutase(SOD), malondialdehyde(MDA),cell apoptosis and morphology, which can provide basis for treatment of osteoarthritis in clinical application of drynariae.Material and method:24 adult New Zealand white rabbits were provided by the experimental animal center of Liaoning University of Traditional Chinese Medicine,were randomly divided into normal control group,model control group,the treatment group and control group,each of 6 dimensional force only. In the left hind limb extension tubetype plaster cast fixation(6 weeks),resulting in the rabbit kneeosteoarthritis model. After modeling,Rhizoma drynariae group according to the dose of 1.5g/Kg/d was given by intragastric administrationof Rhizoma Drynariae,dimensional force group at a dose of 7Omg/Kg/d given orallydimensional force,normal control group and model control group the same dose of normal saline.After 4 weeks of continuous treatment,ear venous air embolismmethod,all animals were sacrificed,surgical operation in eachrabbitsleft posterior knee articular cartilage,synovial membraneseparation.Part fixed in formalin solution,to observe the changes of histology and morphology of HE staining,apoptosis ofsynovial cells was detected by TUNEL;a part of Trizol solution,4 degrees Celsius refrigerated,using RT-PCR(reverse transcriptionpolymerase chain reaction)method for the detection of MMP-3,TNF-α,IL-6;another part of the ground,homogenate aftercentrifugation(3500r/min)10min,SOD(with the supernatant wasdetected by xanthine oxidase),MDA(thiobarbituric acid method).Results: 1.Compared with the normal group,the expression levels of MMP-3m RNA,TNF-αm RNA, IL-6 m RNA levels were significantly increased, thedifferencewas significant(P<0.01).The two dimensional force group,MMP-3m RNA,TNF-αm RNA,and IL-6 m RNA compared with the model group, the expression level was significantly decreased,with statistical significance(P<0.01).The two dimensionalforce group,MMP-3m RNA,TNF-αm RNA,and IL-6 m RNA expression level between,there are obvious differences(P<0.01).2.Compared with the normal group,model group,MDA content of eachactivity of SOD increased significantly, the two dimensional force group and SOD activity were decreased,the difference was significant(P<0.01).Compared with the model group,the group and the dimensional force group,SOD activity,MDA content decreased significantly,the difference was significant(P<0.01). Comparisonof Rhizoma Drynariae group and dimensional force group, SOD activity, MDA content,the difference was statistically significant(P<0.01).3.The extent of lesion morphology of Rhizoma Drynariae group,dimensional force group rabbit synovial significantly lighter than the model group compared with normal group, but there is still a certain degree of difference, and Rhizoma Drynariae group were significantly lighter and external force group.4.Apoptosis of TUNEL rabbit synovial tissue staining was positivein normal group,only weak positive expression in model group,thenumber of synovial cells,there was significant differencecompared with the normal group(P<O.01). The two dimensional force group and synovial cells were more,higher than that in the normal group(P<O.01),but significantly lower than the model group,the difference was statistically significant(P<O.01).Comparison ofthe two dimensional force,between groups,there was a significant difference(P<O.01).Conclusion: 1.Expression of Rhizoma Drynariae can inhibit synovial MMP-3,TNF- α,IL-6,promote the recovery of themorphology,reduce thedamage to the body of free radicals,which is effective in the treatment of knee osteoarthritis.2.Rhizoma Drynariae can effectively inhibit the apoptosis of synoviocytes in articular cartilage,delaying the progression of the disease.3.The control effect is obvious on knee osteoarthritis, the treatment effect is better than that of dimension.
