Objective: Traditional chinese medicine analgesia Babu paste that was to research analgesia effects in mice for traditional chinese medicine analgesia Babu paste by establishing mice pain model(hot plate method experimental model and mice tails pressure method experimental model;to research bone cancer pain’ effects on mice behavior and traditional chinese medicine analgesia Babu paste analgesia effects on cancer pain mice by injecting LLC into C57BL/6 mice tibia bone marrow cavity to establish tibia cancer pain model and apply externally traditional chinese medicine analgesia Babu paste;to research addiction in mice after using traditional chinese medicine analgesia Babu paste by establishing jumping mice model.The author provide data for pain experimental research in order to explore the analgesic effort of traditional chinese medicine analgesia Babu paste, and provide scientific basis for medications for clinical treatment of pain. Materials and methods: 1.Materials The SPF grade animal laboratory, SPF grade Kunming mice, C57 BL / 6 mice, Lewis cell lines, hot plate measuring pain instrument, rat tails tenderness instrument, electronic scales, constant temperature water bath box, OlympusBX51 microscope, thermal radiation stimulation equipment, von Frey cilia, cell counting board, nitrogen, domestic desktop centrifuge, traditional chinese medicine analgesia Babu paste,Voltaren cream, 6% sodium sulfide solution, naloxone hydrochloride injection, penicillin- streptomycin, DMSO, FBS, D- Hanks solution, DMEM high glucose medium. 2.Methods Through hot plate method and put the mice on hot plate to determine changes of mice pain threshold in groups before and after treatment. Through tail pain tenderness and place the mice tail on the tail pain tenderness instrument to determine changes of mice pain threshold in groups before and after treatment,and then to evaluate analgesic effect for traditional chinese medicine analgesia Babu paste;establish bone cancer pain in mice model by referring to schweiMJ and improved method,and observe the mice general condition(such as diet condition, weight change) in each group,and have pain behavioral tdetection on mice in each group(such as mechanical allodynia testing, thermal stimulation threshold detection, spontaneous pain behavioral tests) and radiographic observation.;By jumping test in mice to determine the paste addiction Results: 1.Experimental study on hot plate test for analgesia effects on Babu paste:mice weight and pain threshold in each group before treatment were normally distributed,comparison between the two groups showed no significant difference(P>0.05).Comparison in pain threshold difference in each group before and after treatment :The blank group was compared with the matrix,which was no statistically significant P = 0.98> 0.05,the blank group was compared with each Chinese medicine dose group and positive control group,which was significantly different P <0.05;It was found that increased dose level was closely related to the size of dosage by the pain threshold difference comparison between dose 1, dose 2, dose 3 and 4 dose groups before and after treatment. 2.Rat tail pressure experimental study for analgesia effects on Babu paste:Comparison in pain threshold difference in each group before and after treatment:There was no significant difference between the control group and the matrix group(P> 0.05),The blank group was compared with each chinese medicine dose group and positive control group,which was significantly different( P <0.05);Pain threshold difference comparison in each group:Dose group 4> 3 dose> dose group 2> 1 dose group,and increased dose level was closely related to the size of dosage. 3.Experimental of bone cancer pain,Mice showed hyperalgesia in the study of pain behavior,which was manifested by pain aggravation with the increase of time.Measurement and comparison mice pain threshold in each group:Analgesic effects with increasing doses of the chinese medicine in each dose group was enhanced. 4.Jumping test in mice experiments:No significant difference between the blank group, the matrix group and each chinese medicine group.The positive control group compared with each group.which was significantly difference(P <0.05). Conclusion: 1.Traditional chinese medicine analgesia Babu paste has a better analgesia effect Analgesic experiments of hot plate and rat tail pressure methods for traditional chinese medicine analgesia Babu paste:Analgesic effects comparison in each group:dose group 4>dose group 3>dose group 2>dose group 1> blank group=matrix group. 2.Analgesic effect on traditional chinese medicine analgesia Babu paste has dose correlation. 3.Traditional chinese medicine analgesia Babu paste has analgesic effect on rat model of bone cancer pain,and analgesic action depends on the size of dose level. 4.The use of traditional chinese medicine analgesia Babu paste is safe and non-addictive.
