Objective:Purpose of the study is to detect the concentration of TNF-α and IL-1 in the synovial fluid of PTA. To observe the relationships between both TNF-α 、 IL-1 and knee post-traumatic arthritis X-stage( Kellgren & Lawrence criteria). To investigate the internal relationships between inflammatory cytokines and the severity of knee PTA,and to search biochemical indicators to reflect the degree of PTA.Material and method:We selected randomly 40 patients(21 males and 19 females,18-30 age)with knee PTA from our outpatients and inpatient in 2014(since January to June). The patients according to X- stage(Kellgren &Lawrence criteria) were divided into 2 groups(light and heavy),20 patients in each group,extract effusion of knee joint,measured TNF-α and IL-1 potency.In addition to randomly select 10 healthy volunteers(no other knee joint disease) the same extraction knee joint fluid inspection as control, Of which 6 were male,4 female, age 19-32 years old, average age is 24.59 years old l. Adopt enzyme-linked immunosorbent assay(ELISA) was detected TNF-α and IL-1. Applying SPSSl6.0statistical analysis software, t test, the concentration of experimental observation of TNF-α and IL-1 in each group has no statistical difference. And using Pearson correlation analysis to analyze the relationship between concentration and PTA X-ray grading of TNF –α and IL-1.Result:Liquid TNF- α and IL-1 concentrations of knee traumaticarthritis were higher than those in the control group, the difference was significant(P < 0.05).Each group of knee traumatic arthritis TNF- α and IL-1 concentration with X-ray stage increased, and there were significant differences(P<0.05).Conclusion:Aggravated knee traumatic arthritis synovial fluid of patients with TNF-α and IL-1concentration increased gradually with the disease. X-ray concentration and PTA TNF-α and IL-1 staging is closely related to the severity of the concentration of PTA, suggesting that TNF-α and IL-1 are closely related to the alpha and. To detect the concentration of TNF- α and IL-1 can be used as an indicator to judge the degree of joint lesions.
