This thesis reported the studies on hepatoprotective and antihyperlipidemic activitiest of effective extractives from medicinal plant Desmodium Sambuse D C.The hepatoprotective and antihyperlipidemic activities in D.Sambuse were studied by using the experiment of protecting liver, lipid lowering and antioxidant activity. 1) The mouse model of acute hepatic injury was established by giving CCl4 abdominally to test the hepatoprotection activity of different extracts from D.Sambuse. The results showed that all parts significantly reduced the activities of ALT and AST in serum and the levels of MDA in liver tissue, and raised the levels of SOD in liver tissue markedly, 40% ethanol extract exhibited a remarkable protective effect on acute hepatic injury induced by CCl4 in rats. 2) The mouse model of hyperlipidemia was established by lavaging high-fat emulsion to test the lipid-lowering activity of different extracts from D.Sambuse. The results demonstrated that all parts significantly reduced the activities of TC and raised the levels of SOD in liver tissue, high and low dose group of 40% ethanol extract could reduce the levels of LDLC markedly. 3) The antioxidant activity of different extracts from D.Sambuse were evaluated by DPPH and ABTS assay in vitro. The order of antioxidant activity in DPPH and ABTS assay were 40%>20%>60%>80%>100% and 40%>20%>60%>100%>80% ethanol extract, moreover, there was a positive correlation between the antioxidant capacity and the content of ethanol extracts.In earlier studies the flavonoids and polyphenols were obtained from D.Sambuse, there may be a positive correlation between the content of flavonoids, polyphenols and hepatoprotective, lipid-lowering, antioxidant activity, so the content of flavonoids and polyphenols in different extracts of D.Sambuse. were determined. The determination of polyphenols in D.Sambuse. was established with ultraviolet spectrophotometry in 734 nm and gallic acid was used as reference material, the content of polyphenols in different extracts of D.Sambuse was determined by Folin-Ciocateau colorimetry. The result was: 40% >20% > 60% >water >100%>80%. The flavonoids contents were determined by the way of Na NO2- Al(NO3)3-Na OHThe order of flavonoids contents was: 40%>20%> 60% >100%>80%>water, both of the results verified to be a positive correlation between hepatoprotective, lipid-lowering, antioxidant activity and the content of flavonoids and polyphenols.40% ethanol extract was confirm effective extractive by studying and comparing biological activities of diffirent ethanol extracts. Chromatographic techniques were applied to isolate and purify the constituents in 40% ethanol extracts with silic gel, column, reverse phase silic gel, MCI gel, Sephadex LH-20, macroporous resin and polamide. The structures were elucidated by spectroscopic methods. 14 compounds were isolated from effective part, in which 11 compounds were identified as: 1,2- lycerol dioctadecenedioic acid(JP-1), β-amyrin(JP-2), betulic acid(JP-3), β-daucosterol(JP-4), o-hydroxy benzoic acid(JP-5), β-sitosterol(JP-6), luteolin(JP-7), d-catechin(JP-8), epigallocatechin(JP-9), luteolin-4′-O-β-Dglucopyranoside(14), apigenin-7-β-D-glucoside-(1→2)-β-D- glucoside(JP-15). JP-1, JP-14 were obtainded from the plant firstly. The antioxidant activity of the compounds was tested using DPPH and ABTS assay, and the antioxidant capacity of the luteolin(JP-7) was much higher than other compounds and the reference(BHT, PG and BHA).
