【Objective】 To explore the role of miR-194 in proliferation,migration, invasion an-d the epithelial–mesenchymal transition of human basal-like breast cancer cell line HC C1937.【Methods】There were 3 groups,i.e. experimental group HCC1937 with lentivirus-m-iR-194-EGFP(MOI=50), negative control group HCC1937 with lentivirus-CON157-E GFP, and blank control group HCC1937 without lentivirus.miR-194 expressions were de tected by real-time quantitative reverse transcription-polymerase chain reaction(q RT-PC R);cell growth ability of HCC1937 was determined by Cell Counting Kit-8 assay; cell mi gration and invas-ion ability of HCC1937 were measured by a Transwell assay, and the pr otein of epithelial-mesenchymal transition phenotype was measured by Western blot.【Resμlts 】The expression of miR-194 in experimental group(432.93±0.01) was h ig-her than negative control group(0.90±0.02) and blank control group(1),P=0.000; ex peri-mental group cells(44.81±0.11%) grew more slowly than negative control group cells(104.29±0.13%) and blank control group cells(100±0%), the difference was st atistica-lly significant(P =0.000); cells number of migration and invasion were both lowe r than neg-ative control group and blank control group, the difference was statistically sig nificant(P<0.05); Western Blot showed that the expression of E-cadherin(experiment al group:0.175±0.053, negative control group:0.516±0.032,blank control group:0.433±0.047)decreased obvi-ously, P=0.017,while the expression of N- cadherin(experimental gr oup:0.342±0.012,neg-ative control group:0.137±0.007, blank control group:0.142±0.021)i ncreased obviously, P<0.05, the difference were statistically significant.【Conclusion】 miR- 194 can inhibit basal-like breast cancer cells(HCC1937) prol if-eration, migration and invasion, and also affect EMT process in HCC1937.
