Objective: This experiment is desired to establish a compound model of chronic periodontitis and vascular calcification, so as to study the relationship between periodontitis and vascular calcification.Methods:Forty male Wistar rats were randomly divided into four groups:control group(group C), periodontitis group(group CP),vascular calcification group(group VDN),compound group(group CP+VDN).Every group accepted the corresponding manages to establish the animal model. Eight weeks later, all the rats were sacrificed and the following items were observed: alveolar bone loss and histological examination of periodontal tissue were taken to test the establishment of periodontal model; calcium assay, alkaline phosphatase activity; expression of mineral related factors and Von Kossa staining of vascular tissue were taken to test the establishment of vascular calcification; inflammatory factors and plasma lipids in serum were tested at the.Results: Through detection of periodontal tissue, serum and vascular tissue, animal models were successful. Histopathologic observation revealed: obvious inflammation of periodontal tissue was obversed in group CP and CP+VDN. alveolar bone resorption in group VDN+CP and CP was higher than that in group C and VDN(P<0.05).,and that in group VDN was higher than group C. Vascular calcification lesions of aorta were formed in group VDN and CP+VDN. The calcium assay, alkaline phosphatase activity, red Mineralized nodules deposition in group VDN and CP+VDN were higher than in group C and CP(P<0.05), and that in group CP+VDN was significantly higher than in group VDN(P<0.05); the level of TC, LDL-L in group CP+VDN and VDN was higher than that in group C and CP(P<0.05), and that in group CP+VDN was significantly higher than in group VDN(P<0.05). Animals in group CP+VDN and CP showed higher level of CRP, PGE2, IL-6, TNF-α in serum than that in group VDN and C. The level of HDL-L in group C and CP were significantly higher than the other groups. Animals in group CP+VDN and VDN showed higher expression of OPN, BSP and Cbfa1 in that in group CP and C, the expression of these mineral factors in group VDN+CP was higher than in group VDN.Couclusion:The result showed that the expression of the above items of periodontitis and vascular rats were higher in VDN+CP rats than that of periodontitis rats or vascular rats. The results promted that periodontitis have some promoting effect on vascular calcification; vascular calcification might have some promoting effect on periodontitis.
