| Objective: To test and analyse the special troops’(including submarine crew, the pilot) ACE gene polymorphism, clarify the special characteristics of ACE gene polymorphism, and provide basis for selecting special high-quality warfighters such as submarine crew, pilot etc. And to analyze the genotyping and allele frequency of ACE gene in healthy people and patients with essential hypertension(EH), explore its correlation with EH disease.Methods: 170 of commando brigade(submarine crew, the pilot, etc.) blood samples, 246 of EH patients blood specimens and 500 blood samples of healthy people were collected as the research objects, and detailed information of all specimens was recorded, such as name, age, domicile of origin, nationality, blood pressure, family history, training and psychological test results. PCR technique was used to detect the insertion/deletion polymorphism of 287 position in exon 16 of ACE gene, and a number of biochemical tests, serum ACE and plasma Ang Ⅱ level were detected in some specimens.Results:(1) The frequency of I allele of ACE gene was significantly higher than D allel in the special forces soldiers, and the frequency of ACE gene type II of special forces soldiers was significantly higher than healthy people; the superiority rate of physical training performance of special forces soldiers with type II ACE gene was obviously higher than that of other genotypes.(2) In EH group, the serum ACE levels of patients with II, ID and DD genotype were(36.69 + 14.05) U/L,(42.98-16.61) U/L and(49.37 + 17.43) U/L, respectively, with the highest serum ACE levels detected in DD genotype subgroup, the difference was statistically significant between groups(P< 0.05). There was no statistically difference in the plasma Ang Ⅱ levels of three genotype groups(P> 0.05);(3) As the increasing of hypertension grading, serum ACE and plasma Ang Ⅱ level gradually increased, the difference had statistical significance(P< 0.05). The serum ACE level of EH patients are correlated with ACE gene polymorphisms(r = 0.324, P< 0.05).Conclusion:(1) The ACE-I allele could be a genetic basis for the selection of high quality special forces team, and can be applied to force the establishment of the standard of physical training.(2) The ACE gene polymorphism and serological levels are associated with the occurrence of hypertension, and those can be used to select special forces soldiers to reduce attrition, improve the battle effectiveness. |
