Self-incompatibility represents the most widespread strategy employed by plants, which enables the pistil of a flower to recognize and reject self-pollen or pollen from genetically related individuals, thereby preventing inbreeding and promoting outcrossing. Compositae are generally self-incompatible. Using self-incompable lines to produce crossing breed is an necessary method for plants, however, identifieation of self-incompatibility is a basic step.It is important to accelebrate pace of breeding proeess. In this experiment, on zinnia preliminary study of pollination biology, artificial and natural pollination in field tests and the use of fluorescence microscopy of the pollen and pistil interaction observation, according to a certain index standards for Zinnia self-incompatibility in inbred lines identified to determine their level of self-incompatibility, and research to overcome self-incompatibility of the method, so as to zinnia inbred, hybrid study provides a theoretical basis and practical significance.The results are as follows:(1) Pollination biology By emasculation not bagging, bagging and emasculation not bagging male experimental verification zinnia natural conditions in the basic reproduction. The results showed that: zinnia under natural conditions can a small amount of cross-pollinate fruit, in the absence of pollinators in the case of self-pollination is very low, much lower than the rate of artificial pollination, indicating zinnia of self-incompatibility and natural conditions of the mainly self-incompatible flowers, in the production of a large number of inbred lines to obtain seeds of the case require artificial pollination.The P/O value of the determination, that the breeding system of zinnia mainly facultative outcrossing type.(2)Identify of self-incompatibility To flower artificial pollination of self-compatible and the index as the main standard, secondary reference to the natural self-compatibility index and bagged under the fluorescent microscope observation, the following tentative standard self-compatibility zinnia: Flowering self-compatibility index is less than 0.2 as a self-incompatibility system; flowering self-compatibility index greater than 0.2 less than 0.5 Positioning weak self-incompatibility system; flowering self-compatibility index greater than 0.5 less than 0.7 as self-compatible and the Department of flowering self-compatibility index of 0.7 or more for the high self-compatibility system of self. The light of this standard can be drawn:strong self-incompatibility system as J17 and J9; Weak self-incompatibility system as:J7J,J14G,AP,J7N and J11; Self-compatible and lines for the A1-GH,A2,A4,S5,J6,J8,J13,J14,J15,J18; High self-compatible, and lines for the A3 and J10.(3) Pollinated by different periods and different pollination methods, and different concentrations of reagents on zinnia overcome the effect of self-incompatibility. The results showed that: bud pollination can overcome self-incompatibility zinnia; different pollination methods of study, with strains of different flowers, as well as fellow brothers and sisters all have better pollination seed set; different reagents,10% sucrose solution treatment has good effect; 2% and 3% NaCl solution treatment in improving the self-incompatibility of Zinnia elegans has a role; but 0.05%,0.1%,0.2% of the boric acid treatment results were very close.
