| Tea (Camellia sinensis(L.)O.Kuntze)Theaceae Camellia,is the important economic crops originated from China.It has important significance in establishing the mechanism of embryo tea regeneration system by researching . The Inducing condition and mechanism of somatic embryogenesis.On aiming at low rate of tea body embryo and differentiation,this paper improved the rate of tea body induction by selecting the foreign implant reasonablely and optimizing the training method, while using method of scanning and transmission electron microscope to track the changes of cells in the process of tea body embryo.Based on the process,the inducing embryo contains substances were relately changed, and provided scientific theory for tea somatic embryogenesis and establishing regeneration system.Choosing the tea seeds in maturityE1,E2,E3 and E4 increasing progressively as explant for research.Results showed that the highest rate of inoculation was E4 explant,but lower in inducing somatic embryogenesis,E1 explant had the relative higher consumption rate in the sterilization,and the ability of body embryo induced was not the best.The frequency of somatic embryogenesis differentiation and getting the second body embryos were higher in E2and E3explant,E2best,E3second.The results showed that E2 tea seed explant could become fine materials for body of embryo induced.Using the cotyledon petiole as research material , studied somatic embryogenesis.Results showed that the light training combination of the training in MT + 1 mg/L BA + 0.2 mg/L IBA + 0.05 mg/l 2,4 - D,Light intensity for 500 lx, illumination time 12 h/d.Induction training in MT + 4 mg/L BA + 2 mg/L IBA + 1 mg/L,dark treatment, Polarization in MT + BA4mg IBA2 mg/L,1500 lx,12 h/d, obtained the highest rate of regenerative plants.Using the method of enzyme-linked immunosorbent embryo researched regularity of changing of endogenous hormones ZR, ABA,IAA,GA3 in processing of body indued: IAA,ZR, ABA were basicly consistency,reached peak before the embryo (YM) appeared,the content of GA3was the least before the embryo (YM) appeared (YM) . |
