Compared with the normal diploid rainbow trout,Triploid rainbow trout have fast growth, relatively large body, relatively higher yield of features. For the production of triploid rainbow trout can may avoid decreased meat quality which due to energy transfer and the emergence of marriage during gonad development stage and spawning season, thereby enhancing economic efficiency.Temperature treatment method is utilized to deal with Eggs of rainbow trout forming triploid, which by the retention of the second polar body of fertilized eggs. Compared Rate of triploid eyed and triploid rate by analysis of variance to filter effect of the three factors (treatment temperature, treatment processing time, treatment processing duration).And nucleolar chromosome counting and nucleolar silver staining diploidy detection methods being compared. The conclusions as follows:1.Using heat shock treatment induced triploid rainbow trout and three temperatures all were successfully induced the triploid rainbow trout. As the shock temperature, the death rate of fertilized eggs has increased within 2h. In the 30℃and treatment time lasts longer than 10min conditions rainbow trout eggs all died or stop the growth.So we can considered that very close to 30℃eggs of rainbow trout in the lethal temperature. In other conditions, and as the extension of treatment duration, the death rate of fertilized eggs has increased within 2h.2.Induced by heat shock method, in dealing with the temperature 26℃,the starting time of fertilization treatment after the 15min of fertilization treatment and 10min treatment duration of induction with an induction rate of 65%,for large-scale production of triploid rainbow trout to provide reference basis.3.Nucleolar chromosome counting and nucleolar silver staining all can be more accurate determination of the Triploid induction rate.From the operation perspective, the chromosome counting is relatively more complicated and more time-consuming; and the silver staining method was comparatively simple, rapid, and generally 15 minutes or so.Therefore, carrying out large-scale production of triploid rainbow trout, the most practical approach would be for the nucleolar silver staining.
