Moso bamboo (Phyllostachys edulis), a member of Bambusoideae, Gramineae (Poaceae), was one of the most extensive distribution and highest economy value bamboo used for stuff and food.The effect of NaCl stress on the physiological mechanism of seeds bud period, seedings period and one-year seedlings of Phyllostachys edulis was studied by measuring some physiological index on the condition of different concentrations of NaCl.stress. It was demonstrated that the expressing of PeNHX1 and PeLRR-RLKs which were homologously cloned was affected by salt stress by the analysis of RT-PCR. It was very important for clarifying the mechanism of salt tolerance of Phyllostachys edulis, in terms of physrology, biochemistry and molecular biology. The main conclusions were drawn as follows by the analyses of many indexes:1. Germination percentage and seedlings grew consistently of Phyllostachys edulis seeds had no obviously change in low NaCl concentration (0-50mM), so did physiological index of the trefoil stage. Germination percentage was reduced with the increase of concentration of NaCl in high NaCl concentration (100-150mM). Seeds didn't germinate in 200mM NaCl concentration.2. The physiological index of one-year seedlings of Phyllostachys edulis appeared regular changes during 30 days cultured in 0-100mM NaCl concentration, showing preliminarily that the seedlings had relatively high tolerance to salt stress. Seedlings died in 150mM NaCl concentration.3. After measuring the physiological index of the seeds and seedlings which were cultured under salt stress, the physiological index were made correlation analysis and the conclusion was that the content of proline (Pro), Malondialdehyde (MDA) and chlorophyll (Chls) could be used as important index to measure the damaging extent of Phyllostachys edulis which was under salt stress.4. After measuring the chlorophyll content and chlorophyll fluorescence parameters of the leaves of seedlings, where were cultured under salt stress, Fv/Fm, Yield, qP appeared regularly changes and could be used as important index to measure the damaging extent of Phyllostachys edulis which was under salt stress.5. A gnen which is related to salt stress was cloned and was named as PeLRR-RLKs.6. The result of RT-PCR showed that the expressing of PeNHX1 and PeLRR-RLKs were higher when the seedings were under the condition of salt stress so as to resist stress, it means that the expressing PeNHX1 and PeLRR-RLKs was affected by salt stress.
