| In order to better protect and utilize the mulberry cultivar resources of Hunan Province the purity and authenticity of the main mulberry varieties that are currently planted in Hunan were desiderated to detect. Their genetic relationship and the classification are in needed to determine. It will be the important basis for identification of cultivar and assisted selection for genetic breeding of mulberry in Hunan Province.In this paper the 10 current mulberry varieties in Hunan Province are subjected to study. The process of molecular markers and reaction system for mulberry were established at first. The fingerprints of 10 current mulberry varieties based on Amplified Fragment Length Polymorphism (AFLP) molecular marker were fingered out. The main results are as follows:(1) After a series of experiments the process of molecular marker and reaction system for mulberry leaves were established.(2) The 5 primers were screened from 36 primers, that with abundant bands and high polymorphism. After amplification with the 5 primers to 10 mulberry varieties, altogether 344 polymorphic DNA bands were detected, and the polymorphism rate reached to 27.88%.(3) The coefficient of genetic similarity was 0.79 and the 10 mulberry varieties could be clustered into 3 groups. The triploid cultivar Xiangsang 6 formed a single cluster and the rest 9 varieties formed another cluster, among which Miao 33, a cultivar of Guangdong Mulberry, formed a separate cluster and the rest 8 varieties of Lu Mulberry formed another cluster.(4) The coefficient of genetic similarity between varieties and UPGMA cluster analysis were consistent with the classification based on morphological characters. |
