The Tissue Culture Of Two Maple And Preliminary Research On Prevention Of Tissue Pollution

Acer platanoides'Crimson King'and Acer negundo are colored-leaf trees with ornamental value,widely applies in the city landscape.Both point the city,and purify air.With reproductive problems and high price of woody plants,there is few report about Acer platanoides'Crimson King'and Acer negundo at present.In the study, the Acer platanoides'Crimson King'and Acer negundo are used as explants for tissue culture, preliminary study the factors that influence the rooting of Acer platanoides'Crimson King'and the buds starting optimum medium of Acer negundo .The results show that hormone level ratio, woodiness and sugar concentrations can influence the rooting of Acer platanoides'Crimson King'.In the circumstances that growth in good condition with no lignification , the optimum rooting medium of Acer platanoides'Crimson King'is MS+6-BA0.3mg/L+IBA0.4 mg/L+NAA0.1mg/L,added with sugar 25g/L. Research on the culture of Acer negundo buds starting , the best disinfection time is by dipping the buds in 0.1% mercuric chloride for 5 minutes , and then treating with 75% ethanol for 45 seconds, Acer negundo buds induction medium is MS+NAA0.1mg/L+6-BA1.5mg/L, buds elongation medium is MS +NAA0.1mg/L+6-BA1.5mg/L+GA1.0mg/L. In the tissue culture process of the two maple,there are phenomenons that the tissue culture seedling is polluted. Study on the antibacterial activity of 18 kinds of Chinese herbal medicine to Escherichia coli,Staphylococcus aureus,and Penicillium glaucum.Results show that different concentrations of Rhubarb, Coptis, Chrysanthemum, Berberis and Clove have obvious antibacterial activity, and when PH is six, the Rhubarb's antibacterial activity is the strongest.The five Chinese herbal medicine is used to study the antimicrobial effect to isolated bacteriostasis from tissue culture.In the process of save the polluted seedlings,the high concentration of Chinese herbal medicine can effectively reduce the pollution tissue,but large damage to the tissue culture seedlings. And low concentration while in reducing the rate of pollution is a bit poor, but the damage to the seedlings cultivation is very small.The good thermal stability of three Chinese herbal medicine is constituted in pair.The results show that the antibacterial activity of clove and rhubarb combination is best. Compared to use single antibacterial ,this method obviously enhance the antibacterial activity,and reduce the damage to tissue culture seedlings.
