Moso bamboo Shoots is rich in crude fiber, trace elements and vitamins as well as amino acids, so have high economic value. However, postharvest bamboo shoot rapid increase in cellulose and lignin content significantly affected the nutrition, quality and value of the shoots. In this paper, as materials of bamboo shoots, using homology cloning methods, the genes associated with cellulose synthesis. On this basis, to carry out related cellulose synthesis gene expression in space and time as well as gene expression and study the relationship between cellulose content. Through this study , results as following:(1) Bamboo shoots we are define as the plant material, using homology cloning methods to obtain a complete reading frame containing (ORF) of the four cellulose syntheses'subunit genes and an endoβ-1, 4-glucanase (KORRIGAN) gene.showed that the five genes of Moso bamboo shared high similarity with the CesA genes from Bambusa oldhamii, Oryza sativa, Zea mays, named PeCesA1 (GenBank, GU176303), PeCesA2 (GenBank, GU176304), PeCesA3 (GenBank, GU176305), PeCesA4 (GenBank, GU176306) and PeKOR (GenBank, GU176307).(2) Using semi-quantitative RT-PCR method to detect the gene in bamboo roots stems and leaves; bamboo shoots and upper, middle and base; different dispose for the expression of different times. The results showed that bamboo and bamboo shoots in different parts of the expression of various genes have some differences.High temperature dispose at the gene expression was significantly higher than the expression of low-temperature dispose.(3) Determined using the Mohr-type salt Mao bamboo shoots, middle, base and bamboo roots, stems, leaves the cellulose content. The results showed that bamboo shoots and the upper, middle and base of the cellulose content of about 2.92%, 14.29%, 28.18%, bamboo roots, stems, leaves the cellulose content of about 22.60%, 46.91%, 42.31%. Cellulose content of various parts of the level of gene expression with PeCesA and PeKoR amount of a certain degree of correlation.We cloned four bamboo cellulose syntheses genes and an endoβ-1,4-glucanase gene. It provided a foundation for research of the molecular mechanism of quickly fibrosing after bamboo harvest and also provided a basis for analysing the biosynthetic pathway of bamboo fiber and utilizing genetic resources of bamboo fiber.
