Acacia saligna is a arbor of Mimosoideae Acacia L.,which is introduced from Ethiopia Africa. Materials of the study are seeds,browses and stems of seedling,which are studied about sprouting means and tissue culture of Acacia saligna.Sprouting means of Acacia saligna is studied by testing germination rate with 3 different seed treatments which are etching by concentrated sulfuric acid,dipping in absolute ethyl alcohol and dipping in hot water. The result indicates that imbibition rate, germinability and germination rate are diversely influenced by these three treatments. The imbibition rate of hot-water-dipping is the highest and the 3 levels are up to 100%.The highest germination rate appears when seeds are dipped in 80℃hot water for 40min, which leads to that germination rate is 90% and germinability is 72.15%.The means of etching by concentrated sulfuric acid and dipping in absolute ethyl alcohol have an un conspicuous effect on the indexes.According to variance analysis and multiple comparisons,average germination of dipping in 80℃-water for 40min,has an markedly significant difference with the ones of other treatments.Dipping in 80℃-water for 40min is optimum treatment.Result of Acacia saligna tissue culture shows:1.For Acacia saligna stem,optimum sterilization time is 15s with 75% alcohol and 10min with 0.1% HgCl2,and for browse,optimum sterilization time is 10s with 75% alcohol and 5min with 0.1% HgCl2.2.According to callus growth rate and situation of Acacia saligna,MS is chosen as the optimum callus induction culture medium.3.In the callus induction experiment of browse and stem without axillary buds, growth rate of callus induced by browse is 93.33%,higher than the one by stem which is 81.25%.Both color and texture of callus induced by browse are better than the ones by stem.The result leads to that optimum explant is callus induced by browse.4. When 2,4-D has concentrations of both 1.0 mg/Land 2.0 mg/L,callus growth rate is the highest which is up to 100%,and the color and texture are both good.2,4-D has a restraining effect on fascicle buds differentiated by callus of Acacia saligna.2,4-D is not supposed to be used when inducing callus. 5.When combining 6-BA and NAA,it is found that the highest induction rate, 100%,appears with 6-BA 2.0 mg/L+NAA 0.5 mg/L.It is better to chose MS+6-BA 2.0 mg/L+NAA 0.5 mg/L as culture medium when inducing Acacia saligna callus.6. When inducing axillary buds,it is found that the highest germination rate, 88.7%,appears with MS,and MS is optimum culture medium inducing axillary buds. 6-BA has the best effect on axillary bud germination. Optimum hormone complex inducing axillary bud germination is 6-BA0.4 mg/L with NAA 0.2 mg/L.During the subculture germinated axillary buds on KT at different concentration,it is found that KT at 0.5 mg/L has the best effect on axillary bud elongation.
