In order to clarify the genetic structure of the populations of Apriona germari, and provided the scientific basis for prevention and treatment of Apriona germari, DNA sequences, among six geographic populations of Apriona germari, were amplified by AFLP marker to analyse the genetic differentiation.The research not only revealed the genetic relationship among different populations of Apriona germari, but also provided the scientific basis for discussing the reason of the outbreak of Apriona germari . At the same time, the research could offer the references for improving the technology for monitoring and forecast and formulating the genetic control stragegy. All data was analysed by the software of PopGen 32 to reveal the results:①The establishment of proper AFLP reaction system, which focused on doing research on the genetic diversity of one kind of main pest called Apriona germari from Morus alba, could help analyse the phylogenetic of the pests in the molecular level and open up the new way for pest control. This test used the genomic DNA of Apriona germari as test material. In order to optimize the reaction system, different concentration of Mg2+, dNTP, primers and different dosages of Taq DNA polymerase in the pre-amplification system, and the dilute multiple of pre-amplification production, different concentration of dNTP, primers, and Mg2+ and different dosages of Taq DNA polymerase in the selective amplification system were tested by single factor tests. It was proved that the optimized system of AFLP for Apriona germari was built. The pre-amplification system included Mg2+ 15.0×10-4 mol/L, dNTP 15.0×10-5 mol/L, primers 10.0×10-7 mol/L, and Taq DNA polymerase 1 U. The selective amplification systerm included 40 dilute multiple for pre-amplification production, dNTP15.0×10-5 mol/L, primers 5.0×10-7 mol/L, Mg2+ 15.0×10-4mol/L,Taq DNA polymerase 1 U. The AFLP map of Apriona germari, with clear strips, could be gotten by the optimized reaction system.②DNA sequences, among six geographic populations of Apriona germari, were amplified by AFLP marker to analyse the genetic differentiation. It was shown that, by the result of this research, 241 bands were got with 5 pairs of primers amplifying the sequence, including 196 polymorphic bands, and the average polymorphism accounted for 81.33%, with 13 specific bands from Hangzhou in Zhejiang province. It could be seen that, by the result of the genetic distance, the average genetic distance of the four geographic populations from Hebei was 0.270 3; the one between Hangzhou in Zhejiang and Jiyuan in Henan was 0.397 2; the average genetic distance between Zhejiang and Hebei was 0.4182, and the one between Hannan and Hebei was 0.3334. The dendrogram also indicated that the farther the geographic distance between two populations was, the more significant the degree of genetic differentiation was.
