Dendrolinus punctatus tabulaeformis Tsai et Liu is one of the pest seriously jeopardizing Pinus tabulaeforis Carr. This text selects the Dendrolinus punctatus tabulaeformis Tsai et Liu of different stand types as object and research the variation discipline of digestive enzyme series and detoxification enzyme series inside the intestine of the Dendrolinus punctatus tabulaeformis Tsai et Liu after feeding on different types of stand. Using the method of molecular markers determine the partial gene sequence of mitochondria cytochrome C, in order to probe the relationship between genetic differentiation of Dendrolinus punctatus tabulaeformis Tsai et Liu populations and types of stand, aiming at through the two aspects research of physiological and biochemical and molecular biological of Dendrolinus punctatus tabulaeformis Tsai et Liu to reveal the mechanism and response of Dendrolinus punctatus tabulaeformis Tsai et Liu on mechanism Pinus tabulaeforis Carr under effect of different types of stand for providing the scientific basis of researching the coevolution between Dendrolinus punctatus tabulaeformis Tsai et Liu and Pinus tabulaeforis Carr under the environment duress and integrated control systems.The effort of feeding on Pinus tabulaeforis Carr of different types of stand on the midgut digestive enzymes is determined. The results show that: after feeding on Pinus tabulaeforis Carr from different types of stand, the highest proteases activity of Dendrolinus punctatus tabulaeformis Tsai et Liu is found in pure forest (5.132 U/mg) which is significantly higher than from other mixed forests, in addition manifesting there are significant different between it and Dendrolinus punctatus tabulaeformis Tsai et Liu from others plots (P<0.01);the trend of lipase activity is the Dendrolinus punctatus tabulaeformis Tsai et Liu lipase from pure forest is higher than from other mixed forests which is 0.171U/mg, and showing significant different on the 5% level; however there are no significant different between Dendrolinus punctatus tabulaeformis Tsai et Liu lipase from Aspen and Pinus tabulaeforis Carr mixed forest and Larch and Pinus tabulaeforis Carr mixed forest, the Dendrolinus punctatus tabulaeformis Tsai et Liu lipase from Apricot Pinus tabulaeforis Carr mixed forest is the lowest which is 0.089U/mg and having significant different with other plots, there is no significant different between amylase activity of Chinese caterpillar from different plots.The effort of feeding on Pinus tabulaeforis Carr of different types of stand on the body's detoxification enzyme is determined. The results show that after feeding on Pinus tabulaeforis Carr from different types of stand, the highest body's carboxylesterase activity of Dendrolinus punctatus tabulaeformis Tsai et Liu is found in pure forest (0.083 uMmg-1Pr.min-1), having significant different between it and carboxylesterase of Dendrolinus punctatus tabulaeformis Tsai et Liu from others plots, while there is no significant different between enzyme activity of Dendrolinus punctatus tabulaeformis Tsai et Liu from pure forest and Larch Chinese mixed forest,the lowest enzyme activity is found in Dendrolinus punctatus tabulaeformis Tsai et Liu from Apricot Pinus tabulaeforis Carr mixed forest which is 0.033 uMmg-1Pr.min-1 and has significant different with other plots; the trend of Glutathione-s-transferase activity represent that the enteral activity of Dendrolinus punctatus tabulaeformis Tsai et Liu from pure forest is the highest which is 0.539U/g prot and has significant different with other plots followed by the activity of Dendrolinus punctatus tabulaeformis Tsai et Liu from pure forest which is 0.447U/g prot, the lowest activity is found in Dendrolinus punctatus tabulaeformis Tsai et Liu from Larch Pinus tabulaeforis Carr mixed forest and Apricot Pinus tabulaeforis Carr mixed forest and having extremely significant different with other plot(sP<0.01), there is no significant different between acetylcholine activity of Dendrolinus punctatus tabulaeformis Tsai et Liu feeding on pine needles from different plots.Research population differentiation of Dendrolinus tabulaeformis punctatus Tsai et Liu. The results indicate that in the gene Sequence of 544bp, the average contents of A, T, C and G were 30.6%, 39.4%, 15.3% and 14.7%, respectively, and the contents of A+T 70% were obviously higher than that of C+G 30%, which manifest an obvious deflection. The 544bp exhibits an A/T bias across all sites. 64 nucleotide sites show variation in this sequence fragment and the variability is 11.7%. Nucleotide sequences in genetic distance are 0.002-0.046, indicating a low genetic variation. The dendrogram was constructed by using NJ and MP method, and the results indicated that the genetic differentiations are correlated with the types of stand. Further research shows that the coefficients FST of gene differentiation are between -0.128 to 0.117, the number of migrant per generation all greater than 1, manifesting that there are some degree of genetic differentiation and gene exchange between Dendrolinus punctatus tabulaeformis Tsai et Liu populations from different types of stand.
