This paper systematically reseached tissue culture and softwood cuttings of Nitraria sibirica Pall. superior, including the sterilization of explants, subculture of aseptic seedling,rooting and softwood cuttings. And through studying the influence on the electrical impedance of Ulmus pumila family under alkaline-salt tolerance, it provided a new basis for researching alkaline-salt and its related indicators of Ulmus pumila.The main results were as follows:1. The suitable disinfection of collected explants was directly brought into the super clean table. Firstly they were soaked with 70% alcohol. After 25s, collected explants were disinfected 8 minite with 0.1% HgCl2 The lowest contamination rates of the four clones were 25.00%,28.57%,24.00%,26.92%.2. Study on the effect of 6-BA, KT, NAA, IBA to the proliferation of vitro,6-BA and IBA were suitable plant growth regulators to the proliferation of four clones.3. N6 medium was more suitable for cultivation than MS, B5, WPM subculture. The best ratio of subculture on Shuanggang01,Shuanggang02 were BA 1.5mg/L+IBA 0.5 mg/L; Shuanggang 03's ratio of mitogen and growth hormone in the subculture BA 1.0mg/L+IBA 0.5 mg/L; the best ratio of mitogen and auxin of Shuanggang 04's subculture was BA 1.Omg/L+IBA 0.2 mg/L.4.The best mediums for rooting of four clones were 1/2MS+NAA 0.5 mg/L.5. Effects on the rooting rate and rooting number was extremely obvious among the Nitraria sibirica clones;it did not affect to the main roots'length.There is remarkable influence on rooting rate to the hormone density, the process time and the hormone type, but the opposite on rooting number and main roots length. Shuanggang 04 had the best rooting effect. It takes the best combine with soaking 2h in 250 mg/L Genduozhuang.6. With the increasing concentration of alkaline-salt, extracelluar resistance and relaxation time of all families initially decreased, then increased, and then finally decreased again. Intracellular resistance of families 2,6 and 9 revealed a trend of the "M".7. Distribution coefficient of relaxation time presented irregular change. Extremely significant positive correlations existed among Re,Ri andτ. Extremely significant negative correlations existed between(?) and Ri,τ,Pn. z with Pn presented significant correlation. Extracellular resistance and relaxation time could effectively represent Ulmus pumila's tolerance of alkaline-salt stress resistance.
