Chrysanthemum morifolium 'Huoyan' is commonly use for viewing and ornamental gardens. The features are fast propagation, more flower types, more colors, resistant to strong and ornamental value. In this paper, SQU gene from Lotus japonicas is selected as transformation gene. Hygromycin was used to screen, and agrobacterium was used to transfer into Chrysanthemum morifolium'Huoyan'. Efficient and stable genetic transformation system has been explored. New varieties which have unique petals style are developed and the influence of phenotype of Chrysanthemum morifolium'Huoyan'by SQU gene is studied, in order that evolution and paternity of SQU gene can be understood and CYC genes resources can be expanded.The following content is the main results of the study:1 Establish in Chrysanthemum morifolium'Huoyan'genetic transformation systemBy analyzing the impact factors in genetic transformation, efficient genetic transformation system has been established.30d Chrysanthemum morifolium explants are used as the material, pre-trained for 2d, infected for 10min with a value of 0.4 OD600 Agrobacterium and cultured for 2d under the conditions of 25℃in the dark. It is through delay training for 4d with inhibitory concentration of 300mg/L of Cef, using selective pressure increments and hyg 8mg/L as initial screening medium. Then the buds which grow based on the conditions of the previous steps are cut into re-selection medium with hyg 10g/L. Finally survival buds was moved to rooting medium, growing robust and produce roots.2 Obtain transgenic plantsHygromycin and gene-specific primers was used to detect transgenic plants. Then three positive transgenic plants were obtained. Genetic transformation efficiency was 1.5‰.3 Phenotype observe of transgenic plantsThrough the experiment of non-transgenic plants, it could find that the plants cultivated in pot which contains soil:Vermiculite=1:1 and trained in Artificial Climate Box for 15d and in house 15d is the efficient way of acclimatization. The transgenic plants were moved in land after acclimatization. By observing phenotype it finds that non-transgenic flowers are lush than transgenic flowers; Non-transgenic inflorescence are radial symmetry, while transgenic inflorescence are incomplete radial symmetry; Deformity of ligulate petals of non-transgenic flowers is 5%, while it of transgenic flowers is 3.3%.
