Spirulirta is a kind of green microalgae which is rich in nutrition and is applied in industrialized production. Selection of spirulina, optimization of carbon, nitrogen and phosphorus in spirulina culture medium, the influence of processing mode and extractin in nutritious materials were studied in this paper, the results of the research are as follows:The screening of spirulina: using the individual size, the specific growth rate and total protein content as evaluation indexs, we got two kinds of spirulina from totle 11 by colligation score: 02 and 13.Effect of carbon, nitrogen and phosphorus on total protein content: the optimal carbon, nitrogen and phosphorus in Spirulina culture medium was obtained by single factor experiments as follows: NaHCO3 7.00 g/L, urea 0.10 g/L, KH2PO4 0.10 g/L. And compare with 02, 13 was better.The optimization of carbon, nitrogen and phosphorus: the optimal carbon, nitrogen and phosphorus in Spirulina culture medium was obtained by orthogonal test as follows: NaHCO3 7.00 g/L, urea 0.10 g/L, KH2PO4 0.10 g/L. Using the optimum combination to cultivate spirulina, the total protein, phycocyanin, allophycocyanin, chromoprotein and carotenoid were significantly higher than comparison.The analysis of carbon, nitrogen and phosphorus'dynamic change and filling material: the change of carbon, nitrogen and phosphorus'concentration had certain rule. Urea was consumed in about one week. NaHCO3 was consumed about 40% in a growth cycle. And KH2PO4 had a little consumption, so we didn't need to add. Therefore, we should replenish material according to the concrete situation.Compare with nutritious materials in different treatment: using four different approaches: fresh algae, oven drying, atmospheric drying and - 20℃freeze drying, there are significant differences among the different treatment of Spirulina in nutritious materials: chromoprotein, algal polysaccharides and carotenoid. The chromoprotein content in atmospheric drying was the highest, and the content of algal polysaccharides and carotenoid in - 20℃freeze drying were the highest.The study on nutritious materials in different extraction: using cold solvent to break the cytoderm, and then extract with water and ethanol, the proportion of nutritious materials were different. The content of phycocyanin in aqueous extract was higher, and the content of allophycocyanin, algal polysaccharides and carotenoid in alcoholic extract were higher. When solvent was removed, carotenoid had the least loss and algal polysaccharides had the most loss in sample without water. All kinds of nutritious materials had much more loss in sample without ethanol, and the most extracted percentage of carotenoid was only 57%.
