Diapause is the common mechanism in insects, which adapts them to circumstances and fundamental events that are critical for the success of their lives. It ensures the survival of insects in unfavorable environments and increases their mating chance through synchronizing the growth rate of populations.Insect diapause mainly by photoperiod and temperature。Generally,long-day and high temperature induced insects into the summer diapause.while,short day and low temperature induced insects into the winter diapause.The onion maggot, Delia antiqua (Diptera: Anthomyiidae), is a serious pest with wide distribution. It makes a great impact on the quantity and quality of the Liliaceae plants. And it has a facultative diapause, which contain winter and summer diapause, entering diapause on pupae.so far,the study about summer diapause of insects is very little,and also comparison about diapause.which mostly make by molecular biology technology. However,the gene is the carrier of genetic information,the proteins are really executor of life activities ,who are products of gene expression - proteins. while the proteome is dynamic. Protein has its own peculiar laws of activities, such as modification of protein processing, transport position, structure formation, metabolism, etc. These are not available from genome research on the informed. Therefore, the complex activities of a comprehensive understanding of life is bound to the protein level in-depth study of organisms. Comparison proteomics research of the onion fly pupae provide more valuable information for a comprehensive understanding of the molecular mechanism of diapause. The initial establishment of the onion fly diapause pupae of two-dimensional electrophoresis system, and the onion fly non-diapause and summer diapause pupae in a comparative analysis and mass spectrometry to try. The main results are as follows.In this study, diapause pupae of the onion fly were used to extracted proteins by two methods,which include cracking, high-speed centrifugation and TCA precipitation.the results show by adding lysis buffer plus liquid nitrogen grinding, high-speed centrifugation than TCA precipitation method more appropriate.the less of proteins loss,and the map clear.Improved by optimizing various conditions,after the initial establishment of the onion fly pupae diapause by the two-dimensional electrophoresis system,we we analysis the map of two-dimensional electrophoresis by 7cm,pH3-10 strips and 12% SDS-PAGE, by image scanning and analysis software was detected in more than 230 protein spots. we found that 1 proteins was only specifically expressed on non-diapaue and 5 proteins were only specifically expressed on summer diapause,7 points have differences in expression levels. For the more significant differential protein spots of non-diapause pupae and summer diapause pupae with MALDI-TOF MS , combined with MASCOT-line retrieval system for comparison.
