Tissue culture is one of the most effective method to deal with propagation of Paeonia suffruticosa Andr.. For Paeonia suffruticosa, tissue culture can not be used in practice because of there being difficulty to rooting, poor quality to rooting during rooting culture, and lower survival rate in seedlings domestication process. So far, the system of tissue culture for Paeonia suffruticosa is still incomplete. In this study, experiments were conducted to establish the sterile system of the three varieties using the bud of Paeonia suffruticosa'ling hua zhan lu','shan hu tai1 and'hong tu'as explants. And then, the effect of improved media that perlite and vermiculite were used to replace agar on rooting culture was carried out based on selecting proliferation media and rooting media. In addition, the endogenous hormone content was measured during rooting the process in vitro. The main results are as follows:(1) Different basic medium and plant growth regulators (PGRs) had great influence on bud proliferation. the most appropriate proliferation medium for 'ling hua zhan lu' was modified WPM+6-BA 1.5mg/L +NAA 0.5 mg/L+Sugar 25 g/L, and the most appropriate proliferation medium for 'shan hu tai' and 'hong tu' was modified WPM+6-BA lmg/L+NAA 0.5 mg/L+Sugar 25 g/L. It was proved that Adding rare earth elements in medium help improve quality of seedlings.(2) The effects of IB A, NAA, basic media and sugar on rooting culture were investigated in this study. The appropriate rooting medium,1/2 modified WPM medium (adding vermiculite:perlite = 1:1)+IBA 15mg/L+Sugar 35g/L for'ling hua zhan lu'was selected out. Rooting rate for the modified medium can reach 90%.(3) Hormone levels changed according to a certain rule during the process of rooting. Zt, GA3, ABA levels decreased with IAA levels increasing before the adventitious roots came out from the cortex of seedlings. Whereas, the levels of IAA,Zt,GA3,ABA decreased and gradually stabilized with the adventitious roots developed after roots take placed.
