WRKY proteins play the most important role in plant. Not only they are involved in metabolic anddevelopmental processes, but also in abiotic stress responses and biotic . Inorder to study functions ofWRKY genes in Capsicum annuum L., 24 WRKY genes were cloned from it. Its amino acids sequence,copy number in genome were initially predicted by initial bio-information method.Main results are gotas follows:1. Based on cucumber genome sequence,57 CsWRKY genes during the incompatible interactionbetween Cucumis sativss L. and Meloidogyne incognita were initially predicted by initialbio-information method.2. Based on the genomic sequence of Cucumis sativss L., using RT-PCR, 24 CsWRKY genes(CsWRKY49,CsWRKY17,CsWRKY37,CsWRKY38,CsWRKY23,CsWRKY24,CsWRKY42,CsWRKY34,46,52,55,56,43,54,47,6,18,5,10,9,14,33,45) were isolated from thecucumber roots inoculated with Meloidogyne incognita, and were located in the 7 chromosomes ofcucumber respectively with blast program.3. The bioinformatic analysis showed that WRKY genes were clustered into three groups.Thephylogenetic analysis showed that the 24 CsWRKY genes were classified into three subfamilies,6(CsWRKY49,CsWRKY17,CsWRKY37,CsWRKY38,CsWRKY23 & CsWRKY24) to group I, andCsWRKY34 to group III, 17 were belonged to group II.24 WRKY genes reacted strongly to nematodesinfections, even in the same group, WRKY genes express differently. That may be the results of genefunction evolution.4. The real-time qPCR revealed that among of the 24 CsWRKY genes, the expression of 14 genes(CsWRKY5,6,9,10,14,23,24,43,45,47,49,37,55,56) did not show significantly differencebetween inoculation and control samples, but for the other 10 genes, there were significantly differentafter root-knot nematodes inoculation, in addition, 8 CsWRKY genes were up-regulated (CsWRKY18,33,34,38,42,46,48,54), and 2 were down-regulated (CsWRKY17,52). The CsWRKY42 were themost significantly up-regulated, after inoculation12 h, the expressed fold was 16 times to 0 h.
