Conservation and utilization of Paeonia delavayi Franch and the identification of distant hybrids are the key technology areas in distant hybridization of P.delavayi. Pre-soaking, treatment with growth regulator and chilling were used to exploration the dormancy and germination of the seeds. Introduction and acclimatization of cultivated seedlings were studied. In the same time, ISSR markers combining with morphological markers were used for the early identification of descendents from distant hybridization of tree peony.1. This study aimed at dormancy-breaking methods of Paeonia delavayi Franch seeds. After 96h-pre-soaking in the water of room temperature the coat-dormancy can be broken. The germination rates of Ow-chilling seeds and 52 w-chilling seeds are higher than other treatments.18℃is the best temperature condition for rooting. Chilling for a period of 8w and lOw after rooting can break the epicotyl dormancy of non-chilling seeds and 52w-chilling seeds. Using ABT and GA3 can break the secondary dormancy. GA3 can break the dormancy rapidly but the seedlings may be weak.2. Acclimation of seedlings has been done in different places. The survival rate of seedlings in Chengde is higher. Growth increment of seedlings in Chengde is higher than other places.3-year-old Seedlings in Chengde can bloom after 3 years.So climate of Chengde is suitble for Paeonia delavayi Franch seedlings. The survival rate of 2-year-old seedlings in Beijing is lower because of the high temperature in summer. Planting in the containers can improve the survival rate but the growth increment is lower.3. Tree peonies from different species are studied to selecting suitable parental combination.The seed maturing rate of Paeonia delavayi Franch hybridizing with Paeonia rockii amount to 87.1%. This rate is higher than Paeonia suffruticosa hybridizing with Paeonia delavayi. Slecting suitable parental combination can be helpful for hybridization of Paeonia delavayi Franch.4. Identification of the hybrids of Paeonia suffruticosa 'Qiu Fa Yi Hao' and Paeonia delavayi has been done using morphological and ISSR markers. The results showed that the F1 hybrids of the parents could be divided into paternal type, maternal type and specific type.12 hybrids have the same results by morphological and ISSR markers, but 4 of the hybrids showed different divisions.
