Stripe rust (Puccinia striiformis Westend f. sp. tritici), caused by Puccinia strriformis Westend f. sp. tritici Eriks is one of the most important disease of wheat (Triticum aestivum L.), which resulted in massive wheat yield loss in several epidemics in China. Due to the special climate condition, Yellow rust spread moderately every 2-3 years in Gansu. Loss of variety resistance to stripe rust pathogene is an important factor causing massive periodical epidemic of stripe rust in wheat production. For probing the disciplinarian of wheat strip rust on pandemic, it is important to research the region relativity on pandemic. High frequency virulent races is SU11 and Hybrid46, among which the main preponderant race CYR33 has led major varieties into susceptible in Gansu. CYR32 took the second place. The minor race was different in different year. Wheat stipe rust belongs to bacterial parasite, the character of heritage is very complex, however, its sexual generation has not been discovered, it couldn't be artificial cultured. The normal indentification methord of races was complex and long, and the accuracy was easily influenced by the indentification condition. Meanwhile, because of the identification limitation, it is hard to differentiate and identify the the type. All these seriously limit the research of biology of population genetic structure.Molecular markers could facilitate the identification and location of novel genes. this research utilizes the RAPD technique to analyse the prevalent race of wheat stripe rust in Gansu. We will establishes the foundation in order to monitor the physiological races of wheat stripe rust and provides a scientific basis for the purpose of resistance breeding and reasonable arrangement.The results of the research as follows:1: The conditions of the RAPD reaction of the wheat stipe rust are optimized and the standard RAPD system, which is appropriate for the Wheat stipe rust is established in Gansu.2: Of the 160 random primers analyzed in this paper are selected, most of which were found out to obtain stable and clear amplification patterns.The RAPD analysis wheat stripe rust in eight major physiological races of the single-spore strains indicates that the heredity and variation among the physiological races of wheat stripe rust in our province is vast, part of genome were still conservative in some degree.3:The RAPD mark of specificity in CYR33,CYR32,Hybrid46-8,shui-4 race four popular were found in the experiment. A pair of specific PCR primers were designed and transformed into the SCAR markers through the cloning, recyling and sequencing of the specific RAPD fragments of CYR33.
