Myostatin (MSTN) as an important cytokine controls growth and development of skeletal muscle in animals. Eight single nucleotide polymorphisms (SNPs) were totally identified in the exonⅠof MSTN gene from 180 (70 of one -year old and 110 of two-year old) large yellow croaker, Pseudosciaena crocea, individuals by PCR-SSCP and sequencing. The eigeht polymorphic sites were located at 28(C→T), 51(T→C), 196(C→T), 264(G→T), 270(G→A), 348(C→T), 349(A→T) and 352(G→A). Base mutations at sites 28, 252 and 349 resulted in amino acid substitutions of L→F, E→K and T→S, respectively. However, other base mutations did not cause amino acid substitution. There were seven genotypes in the examined population, whose frequency was 0.46, 0.17, 0.13, 0.12, 0.07, 0.03 and 0.02 respectively. The frequency of allele A, B, C, D and E is 0.53, 0.34, 0.06, 0.02 and 0.01 respectively. The heterozygosity of exonⅠlocus was 0.60. The PIC was 0.446. Statistic analysis showed that the differences in both genetypes and amino acid sequence had significant effects on the body length and weight of the fish one-year old but not the fish two-year old.Ten single nucleotide polymorphisms (SNPs) were totally identified in the exonⅡo f MSTN gene from 180 (70 of one–year old and 110 of two-year old) large yellow croaker, Pseudosciaena crocea, individuals by PCR-SSCP and sequencing. The ten polymorphic sites were located at 29(A→G), 131(T→A), 206(C→A), 231(T→C), 243(G→A), 261(A→G), 265(T→C), 273(C→T), 297(A→G)and 300(G→A).Base mutations at sites 131, 265, 261, 231, 273, 297, 243 and 300 resulted in amino acid substitutions of L→Q, C→R, S→G, W→S, R→W, T→V, D→N and E→K, respectively. However, base mutations at sites 29 and 206 did not cause amino acid substitution. There were six genotypes in the examined population, whose frequency was 0.57, 0.23, 0.10, 0.04, 0.02and 0.04 respectively. The frequency of allele A, B, C, D and E is 0.75, 0.12, 0.10, 0.03 and 0.01 respectively. The heterozygosity of exonⅡl ocus was 0.33. The PIC was 0.41. Statistic analysis showed that the differences in genetypes and amino acid sequence had significant effects on the body length and weight of the fish.The length of microsatellite sequence is mostly between 64 and 126 bp, and the base mutations are mainly C→T and A→G. 25 single nucleotide polymorphisms (SNPs) were totally identified in the microsatellite sequences of MSTN gene from 180 (70 of one–year old and 110 of two-year old) large yellow croaker, Pseudosciaena crocea.The heterozygosity of microsatellite sequences locus was 0.33. The PIC was 0.31. Statistic analysis showed that the differences in length of the microsatellite sequence had a significant effect on the body length and weight of the fish rather than base mutations of the sequences did.
