Terrestrial cyanobacteria collected from Lanzhou Northern Hill, Minqin of Hexi, Shapotou of Ningxia, Dalate Banner and Balinyou Banner of Inner Mongolia were studied by traditional culture method, genetic polymorphism of 16S rDNA and shape of algal colony on the agar plates, and desertification control technology combination biological soil crusts was explored in the shifting sand dunes of Kubuqi Desert. The primary summaries were the followings:1 Filamentous cyanobacteria Lygnbya, Phormidium and Schizothrix were often dominante genera of biological soil crusts, and they accounted for 86% of the total blue green algae2 For species diversity of cyanobacteria, the highest occurred in loess, then followed by castanozems and aeolian soils, the lowest exhibited in brown caliche soils.3 A new DNA extraction method suit in biological soil crusts of arid area was established. The extracted DNA normally had relative high purity and productivity, so it can be directly used in the latter molecular experiments.4 It was founded that the combination of semi-nested PCR-DGGE could be analyzed genetic polymorphism of cyanobacteria of algal crusts.5 It was discovered that there were obvious differences among colony shapes of different genera on agar, this phenomenon can be used in the cyanobacterial primary classification and count during culture process, and this is a rapid and efficient method to analyze soil cyanobacterial community structure.6 It was discovered that the desertification control technology of combination biological soil crusts was sustainable and efficient method, had wide application foreground.
