The Cross Breeding And Genetic Analysis Of Hybrids Of Pseudosciaena Crocea♀ And Miichthys Miiuy♂

We conduct the cross breeding between Pseudosciaena crocea(?) and Miichthys miiuy(?) and hope hybrid can earn some good characters from its parents. It will be great meaningful in aquaculture and breeding. By the technology of AFLP and SSR to study genetic relationship of hybrid and its parents to show the hybrid hereditary characters.The experiment and results just as follows:1. We cultured the hybrids of Pseudosciaena crocea(?) and Miichthys miiuy(?) successfully. The fertilization rate, the hatching rate and five-month survival rate of hybrids of P.crocea (?)×M.miiuy(?) is 56.25%,45.24%, 0.65% respectively.2. Four primer pairs ( E-AAC/M-CAG E-AAC/M-CAT, E-AAC/M-CTA, E-AGC/M-CTC ) were used to do AFLP analysis for the hybrids and their parents. 312 selectively amplified fragments in total were detected. Among the 312 fragments, 143 female-specific bands and 104 male-specific bands and 32 non-parental bands. There 132 female-specific bands and 18 male-specific bands in the offspring. The similarity index among offspring is 0.891, the similarity index between the offspring and female parent is 0.853, the similarity index between the offspring and maternal parent is 0.271. The genetic distance among offspring is 0.116, the genetic distance between the offspring and female parent is 0.159. the genetic distance between the offspring and maternal parent is 1.307.3. Six primers were used to do microsatellite analysis for the hybrids and their parents. All the loci from the female parent in the hybrid family and no loci from the maternal parent.4. From the AFLP and SSR analysis, most of heredity maternal of the hybrid family from female-parent, but there some gene from maternal-parent. The result showed that the hybrid of P. crocea(?) ×M.miiuy(?) is allogyenesis.
