Chitinases hydrolyle chitin, the second main natural resources and the component of fungal cell walls and peritrophic membrane of insects. Therefore, chitinases are important in a wide variety of biological and biotechnological processes, agricultural and industrial production, natural resources utilization, biological control, environmental protection and medical exploitation. Study on chitinase from soil microbe is important in finding new sources of chitinase using in community.Three strains (A, B and C) producing chitinase were screened in our research from the soil near the chitin factory channel. The strain A belonged to actinomycetes; the strains B and C belonged to bacteria. The strain C which could produce the most chitinase was identified as Stenotrophomonas maltophilia by 16S rDNA method.The research results of fermentation condition of S.maltophilia showed that the most chitinase was synthesized when cultured in the medium containing 1.5% colloid chitin and 1.0% yeast extract for 60 hours (220r/min) by single factor optimal method and uniform design method. The optimal pH and temperature was 7.0 and 30°C, respectively. The crude enzyme was obtained from the fermentation broth by ammonium sulfate precipitation process.
