| Red berry, Myrica rubra Seib.et Zucc, is one of the special fruits as a local famous quality agricultural product in Zhejiang province of China. It is full of Vitamins, trace elements and 17 kinds of amino acid. A novel variety of red berry with better fruit quality and earlier harvest date was found in Xiangshan of Zhejiang, named"Wuzi"red berry. In order to characterize this new variety, the dry and fresh weights, sugar content, titratable acid content, anthocyanin content during fruit development were examined. The enzyme activities related to sugar-metabolism during fruit development were also determined. Molecular characterization based on RAPD markers was used to identify the genetic polymorphism of this new variety. The major results are summarized as follows:1. The dry and fresh weights of"Wuzi"red berry accumulated with fruit development and reached the peak during fruit maturing.2. The sugar and anthocyanin accumulated with the fruit development and rised sharply before two weeks of fruit riping. The major sugar in"Wuzi"red berry was sucrose, which accounted for 2/3 among total sugar in the ripe fruit.3. The content of TA in red berry changed from low to high, then to low during fruit development, and came to peak before two weeks of fruit riping. The sugar content and anthocyanin content were higher in"Wuzi"than in"Dongkui", but the acid content was adverse. The fruit quality of"Wuzi"was better than"Dongkui"based on our analysis.4. The activities of sucrose cleaveage enzymes in"Tanmei"red berry rised during fruit development, while those in"Wuzi"were consistent during fruit development and lower than"Tanmei". The activities of sucrose synthase in"Wuzi"were higher than in"Tanmei". The SPS activity accumulated with the fruit development.5. The activity of FRK was lower than HXK in ripen"Wuzi"fruit, but The activity of FRK was higher than HXK in ripen"Tanmei"fruit.6. Ten varieties, the female parent, two male parents, one to four generations of"Wuzi", local"Tanmei","Shuimei"and"Dongkui"were used to characterize genomic polymorphic and genetic coefficient, respectively. More than one hundred random primers were applied for PCR amplification and 47 primers were selected as better primers. 2403 bands were amplified from the 47 primers, in which 1195 bands were polymorphic. 7. The coefficients among the female parent and one to four generations of"Wuzi"were from 0.94 to 0.98, which were higher than others. The coefficient among"Wuzi"and"Dongkui","Tanmei"and"Shuimei"were lower, only about 0.8. The closest genetic distances were also found among the female parent and one to four generations of"Wuzi".8."Wuzi"red berry was a new variety based on our physiological, biochemical and molecular characterization above. |
