Rotavirus is one of the main pathogeny of causing infant and virous kinds ped-age animals nonbacterial diarrhea. In the developing countries,there are 18 millions children occur midrange and acute diarrhea every year, and the people succumbed to the infection of rotavirus exceed 350000-390000;meanwhile in the developed countries, deprivation of body fluids causing by RV diarrhea is the common etiological factor of children in hospital .According to estimation ,in the global ,the death rate of RV diarrhea is not yet degrade. At the present, duing to the patent medicine is not manufacrured,so the prevention is more important. WHO appealed scitists in countries to develop effective rotavirus vaccine many times, and took it as the one of the most priority developed preceedings. According to estimation , if using the effective vaccine, we can save 500000-1000000 life of children. Vaccine prevention is the optimized selection to control this disease. Along with lucubration of molecular biology, the two main coat protein VP4 and VP7 is not only the base of RV serotype typing, but also the main protective antigen.Accordingly, using genetic engineering, we can express VP4 and VP7 gene as antigen, furthermore in the antigenicity and immunogenicity VP7 is better than VP4, so we choose VP7 as the main researching direction of genetic engineering vaccine of national and international rotavirus. Now. the researching of human rotavirus is further deeply, but the researching of animals esp. pig is in the primary stage.RV is belongs to Reoviridae, the RNA genome contains 11 fragments, each fragment encode a type of protein, 6 structural protein VP1-VP4,VP6,VP7 and 5 non-structural protein NSP1-NSP5. We proceed many researches on the immunologic response of RV infection of human and animals, but the patent protective mechanism is dimness.This research identified that VP4 and VP7 is the main proteantigen on the coat protein, they can stimulate the body to produce homologous neutralization zntibody.In this study, based on the analysis of national and international rotavirus, we can study the molecularly biological characteristic of Rotavirus and lay a solid foundation for the study of new vaccine.
