Study On The ELISA Diagnosis Of Paratuberculosis In Ovine

Paratuberculosis is a chronic and infection disease of cattle, sheep, goat, camelids and other ruminants and nonruminants. The disease is caused by Mycobacterium paratuberculosis and which is chatacterized by chronic diarrhea and emaciation. Johne found this disease in 1895 in the first instance, so paratuberculosis is also named Johne's disease. It was compartmentalized a kind of infection disease B by World Organization for Animal Health and it was compartmentalized a kind of infection disease II in china.Paratuberculosis has a long latent period and clinical signs do not usually occur for lifetime after infection. Untill now, there is untreatable measure for this disease. The only way that prevention and cure is detection, segregation and eliminate the infected animals through selection or contest. Espescially, the study and application of ELISA for diagnosis of paratuberculosis in ovine will have momentous academic and practice significance. Based on these points, the dissertation would lay stress on the study of diagnosis of ovine paratuberculosis disease(OJD) methods.It consists of three chapters:Chapter 1 Paratuberculosis and the basic principle are summarized in this part. Johne's disease is prevalent worldwide and not only causes substantial economic losses to the farming industry, but also potentially threaten the healthiness of human. The technique of ELISA is more and more perfect as time goes by which can detect antigen and antibody.Chapter 2 The study and application of ELISA method for diagnosis of paratuberculosis in ovine are presentation. They are preparated separately that including the immunoaffinity chromatography antigen and horseradish peroxides Conjugated antibody against sheep and goat immunoglobulin G. And the ovine sera detected were absorbed by the antigen of M.phlei which was killed by heating and treated in the ultrasonic wave. ELISA is applicated in ovine sera from six areas. Comparing to regulate diagnosis method including allergic reaction, CF and AGID,we can calculate the result of detection. The percentage of positive sera is 10.8% through the detection of ELISA. And the allergic reaction is 4.9%, CF is 6.6%, AGID is 4.1%.Chapter 3 Above all, we can draw a conclusion that ELISA has a higher sensitivity and specificity than allergic reaction, CF and AGID.
