Cadmium can enter organism through various pathway and cause multiorgan toxicity, including kidney,liver,spleen,lung,cardiovascular,etc.Effects of cadmium on membrane structure and function were further studied in chicken spleenic lymphocyte on the basis of studing on apoptosis mechanisms induced by cadmium in chicken spleenic lymphocyte. In the study, using chicken spleenic lymphocyte as a modle, we detected that the index including LDH content of cellular supernate; composition, content, anti-oxidative function, ATPase activity and fluidity on cytomembrane; [Ca2+]i and CaMmRNA expression in order to reveal toxic effect of cadmium on lymphocyte membrane. The results showed as follows:1.Results of structural analysis of lymphocyte membrane showed that cadmium could induce changes of protein content including increase of zoneâ… ,â…¡,â…¢,â…£, IX, decrease of zone V,â…¥,â…¦; percentage of PC, PE decrease and percentage of PS, SM increase in membrane phospholipid; SA content decrease..All these showed that cadmium could induce disorganization of biomacromolecule in cytomembrane, oxidative damage of membrane protein and membrane phospholipid and cross linkage of membrane protein.2.Results of anti-oxidative function revealed that cadmium could induce the decrease of GSH-Px, SOD activity, the increase of MDA content. This showed that cadmium could induce toxic effect on lymphocyte membrane through oxidative damage of lymphocyte membrane.3.Results of ATPase activity, micro-consistency and fluorescence polarization of lymphocyte membrane, LDH activity in cellular supernate showed that cadmium could induce decrease of ATPase activity, membrane fluidity and increase of membrane permeability.4.By detecting [Ca2+]i and CaMmRNA expression, we found that cadmium could induce disorder of calcium homeostasis and further cause function disorder of lymphocyte membrane by increasing [Ca2+]i and decreasing of CaMmRNA expression.This study revealed toxic effect of cadmium and mechanism on lymphocyte membrane from the subcellsular and molecular level. This will provide evidence for studies on the effect of cadmium on cell, tissular organ and whole organism and establish risk evaluation on the reliable theory.
