Experiment was conducted with Huaguang and Shuguang nectarine cultivars for studying the effects of different plant growth regulation, the base media, carbon resources intensity of illumination, temperature and the dark days on the formation of embryonic callus, During the different periods, observed the state of Huaguang effective callus which were cultured on the chose medium and researched the effects of different media on the callus growth. Observed state and cell structure of callus in the whole process, seek the way of adventitious regeneration.Results are as follows:1. Studied the effects of different plant growth regulation, the base media, carbon resources and environment factors (intensity of illumination, temperature, the dark days)on the formation of embryonic callus: carbon resources influence hardly on inducing embryonic callus, other factors effect remarkably on inducing embryonic callus. During the experiment ,cross cutting back of leaves in both nectarine cultivars cultured on G+2.5mg/l BA+0.5mg/l IBA medium for 21 dark days, then cultured on 1/2MS+2.0 mg/l TDZ+2.0 mg/l BA+0. 5 mg/l NAA ,between 24℃and 27.5℃in the daytime and 2000lx in intensity of illumination, the callus grew stronger and embryoid appeared, differentiated tissue were kept on culture, consequently adventitious shoot from leaves were acquired and the plant regeneration percentage from leaves was 8%.2. In the different time cell structure of callus on the different medium were observed and found: At premier period of the callus growth, the plant growth regulation affected remarkably on the callus growth, the base medium affected strongly on the callus growth later. the callus state in the different time were observed and found: The basic medium mainly effected on the callus state.3. State and cell structure of callus were observed and found: Effective callus which was yellow and white were acquired after explants were cultured darkly for 21 days, then cultured on the medium (1/2MS+2.0 mg/l TDZ+2.0 mg/l BA+0. 5mg/l NAA).
