| The haemolymph and muscle extracts from 7 species of bivalve were tested for agglutinating activity using 10 kinds of erythrocytes of human and animal, 8 species of unicellular algae and 10 kinds of unicellular microbe. The results showed that both haemolymph and muscle extracts of bivalve contain lectins and there were evident difference between them from the same bivalve, and it is conferred that the lectins of haemolymph and muscle extracts from the bivalve were different in types. The erythrocytes from rabbit, Porphyhdium purpureum, Saccharomyces carlsbergensis are the most sensitive cells respectively by synthesizing the agglutination activity of haemolymph and muscle extracts of bivalve.Taking Tegillarca granosa as experiment material, by extraction, fractionation with saturated ammonium sulfate precipitation (40%-20%), gel filtration on the Sephadex G-100 and followed by absorption chromatography on Sepharose 4B, a lectin (TGL-h) was purified from the hemolymph of T. granosa. The muscle lectin of T. granosa (TGL-m) was purified by fractionation of high concentrated ammonium sulfate (65%-45%) , ion exchange chromatography on the DEAE-Sepharose colum and gel chromatography on the Sephacryl S-200-HR colum. Further more, the molecular weighu content of neutral saccharide , pI, ultraviolet absorbing spectrum, amino acid composition of two kinds of lectins were determined, and erythrocytes specificity (native and enzyme-treated erythrocytes) , carbohydrate-binding specificity, thermostability, and influence of pH and salinity,β-mercaptoethanol, bivalent metal cations to the agglutinative activity were compared between TGL-h and TGL-m. They both have the ability of markedly promoting the lymphocyte transform action and the activity of different antibacterial, antifungal. The cross-reactivity of TGL-h and TGL-m was analyzed by double immunodifusion test. It was found that the rabbit specific anti serum against either TGL-h or TGL-m could react with itself, but could not produce precipitation line with the other one, so we draw the conclusion that they had not homology properties to each other.The phagocytosis test on haemocytes of T. granosa in vitro and the effects of... |
