Motilin and ghrelin are important brain-intestine hormone,which regulate functione of stomach and intestine,such as secrete of stomach acid and stomach movement by center and peripheral nerve.Meishan pig motilin and ghrelin genes were cloned by RT-PCR ,and Intronl and of ghrelin, intron2 and intron4 of motilin were amplified from DNA by PCR. Meishan pig motilin and ghrelin genes structure were analyzed by bioinformatiom and biology. Research their Tissue Distribution by semi-quantitative RT-PCR. Chromosome mapping of procine ghrelin and motilin genes by RH. Construct recombinant pDsRed-motilin and pEGFP-ghrelin vectors and their expression in vitro. Research the hormonal response of ghrelin mRNA expression in the IBRS2 cells. Results showed: (1) Meishan pig motilin and ghrelin cDNA had cSNPs. (2) Distribution of pig ghrelin mRNA and motilin mRNA were broad ,among of which Ghrelin mRNA and Motilin mRNA was the best abundance in stomach and small intestine ,while the lowest in liver and in appendices, respectively. (3) Partial genomic fragments of ghrelin and motilin were obtained by PCR of genomic DNA. (4) By radio hybridization mapping, pig ghrelin and motilin were located to pig chromosome 13q31 和 1q21-q27, respectively. (5) Construction combinant pDsRed-motilin and pEGFP-ghrelin vector were sucessful and their expression were highly efficient in RIBS2 cells. (6) Glucose, insulin or T dose-dependently reduced the ghrelin mRNA levels ,but both glucagon and progestin could increase the ghrelin mRNA levels and attenuate their inhibition, respectively. Glucagon and insulin had a conflict effect on glucose transporter (glut4) mRNA levels, glucagon decreased while insulin increased its expression in the IBRS2 cells.
