Porcine reproductive and respiratory syndrome (PRRS) is characterized by reproductivefailureofpregnant sows andrespiratorydistress ofpiglets and growingpigs. It has developedinrecenttwentyyearsandproducedcatastrophiclossesinsomecountries.Itwasreportedthatthis disease had broken on the end of 90s of the last century in China. In order to offer aconvenient tool for PRRS epidemiology survey, evaluation of vaccine immune effect andcustom detection. An immunoperoxidase monolayer assay (IPMA) was developed forantibodydetection of porcine reproductive and respiratorysyndrome virus (PRRSV). Akit ofPRRSV-IPMAwas prepared in the study. The sensitivity, specificity, repeat and expiration ofthekitweretested,andcomparedwithHerdChekIDEXXkit.ThePRRSV-specificantibodiesof experimentally infected pig sera were monitored with PRRSV-IPMA. The results showed50% positive rate(3/6) for the first week sera of post-inoculation , and then obtained 100%positiverate(24/24)fortheserafromsecondtofifthweekofpost-inoculation.Allserafrompigs without inoculation as control group showed 100% negative rate (30/30). In comparison,42serawereusedforparalleldetectionbyPRRSV-IPMAand IDEXXkit.Theagreementratewas 83.3% between two kits. Total 520 sera from thirteen pig farms and 80 sera from sixdisease break farms from different districts of Hei Longjiang, Jilin, Liaoning, Hebei andShanghai were detected with PRRSV-IPMA. PRRSV-antibody positive rate was more than50% in the eight of the thirteen farms, and only two farms was negative. PRRSV antibodypositive rate was from 57.1% to 100% in the six disease break farms. So PRRS disease is asevere harm for pig farms in China. PRRSV-IPMAkit may offer a convenient tool for PRRSepidemiology survey and evaluation of vaccine immune effect. The study have developed aninexpensive, rapid, specific diagnostic assay, which provides a available technique for thediagnosis and serological survey of PRRS and a good further developing commercialantibodydetectionkitforthedisease.
