In this study,according to the particularity of dissolubility protein in variouspathogeny bacterium ,we establish the method of SDS- PAGE for detecting Acidovoraxavenae subsp.citrulli . providing the method of detecting A.a.c for the plant detectdepartmentandport.The testing strains were amplified by polymerase chain reaction using the universalprimers of the bacteria 16S rDNA gene, and the nucleotide sequences of their amplifiedfragment were tested. Analyzed their homology between these sequences and the 16S rDNAsequence of related bacteria strains in Genbank. A phylogenetic tree based on 16SrDNAsequences was constructed,and designed specific primers.The result of PCR was that onlyA.a.c strains came into being eletrophotesis strap.A.a.c was detected from differentpathogenic germs, and a rapid and accurate method for detecting A.a.c was established frommoleculebiology.A specific probe was designed based on difference of 16S rDNA between Acidovoraxavenae subsp.citrulli(A.a.c) and other bacterium,and it was used to detect the testingstrains.The results show that no signal was detected for the others except Acidovorax avenaesubsp.citrulli.ThemethodismorespecificthannormalPCR,anditisalsosensitivemethodtodetectA.a.c.This method can provide a specific, sensitive, rapid detection of A.a.c for quarantinedepartmentandtheseedindustry.
