Avian influenza (AI) is an infectious disease caused by avian influenza virus (AIV), which is one of the most severe diseases for poultry industry. Poultry (chicken, turkey, duck, goose, partridge and so on) and wild birds are all easily infected. In recent years Avian Influenza has resulted in large economic loss, and caused infection and death of human, therefore, it is important in public health.At present detecting methods of AI include virus isolation, serology analysis, molecular diagnosis and electronic microscope technique. However, many methods have the disadvantages of time consuming and/or expensive lab equipments which make them not suitable for clinical instant diagnosis. Gold immuno-chromatography assay (GICA) is a new fast detecting method developing in 1980's. In this paper GICA was successfully introduced to diagnose AI and two methods were established: indirect method to detect antibody against AIV and"sandwich"method with two monoclonal antibodies to detect AIV.1 Establishment of gold immuno-chromatographic assay in rapid detection of antibody against avian influenza virusThe recombinant nucleoprotein of avian influenza virus, expressed by beculovirus expression system, was purified and coated as test line, and the goat-anti-rabbit antibody was coated as control line. Colloidal gold was prepared and labeled with rabbit-anti-chicken IgG. Indirect GICA was established to detect antibody against AIV in chincken serum. If the serum sample contains antibody against AIV red test line will appear; if no specific antibody exists test line will not appear. Red control line will appear in both positive and negative samples. 342 chicken serum samples were detected for antibody against AIV by GICA, hemagglutination inhibition (HI) test and agar gel immunodiffusion (AGID) test, and the results of these three assays were...
