Root-knot nematodes(Meloidogyne spp.) are economically important pathogens of a wide range of crops worldwide.which are causing considerable damage on a variety of crops.Up to date , approximately 81 species of Meloidogyne have been discribed ,of which 30 species have been recorded in China.To clarify the distribution of root-knot nematodes in Hainan Province, four primer pairs (MI-F/R955 , MJ-F/R517 , MI-F/MT-R799 , Me-F/R236 ) was used to identify the species identity of sixteen nematode populations collected from Hainan Province,results revealed that the most of the pathogens are Meloidogyne incognita, M. javanica, M.arenaria and mixed populations of the three, M enterolobii also caused a certain extent damage.M.hapla is a common species,which is widely distributed in the north of china.A single-step PCR diagnostic was developed for M.hapla.The diagnostic primers were designed based on an alignment of mtDNA sequences from M.hapla, M. incognita, M. javanica, M. arenaria and M.enterolobii.The, reliability of the diagnostic test has been validated by screening different geographic populations of five closely related Meloidogyne species and natural mixed soil nematode populations. The test is fast and sensitive, it can be routinely used for direct diagnosis of a single nematode and for detection of M.hapla in mixed soil nematode populations.
